overlay is a casing problem. It doesnt matter when you birth them...you make the choice

Unless your looking to do a casing i'd birth when ready although i'd wait a couple days after i thought it was%100 colonized just too make sure.
It sounds like you got too much in some and not enough in the other so don't forget to shake that baby up between innoculations next time and spread it out some more
