Stem Plugs for cloning?

Sorry if this doesn't go in advanced, but it doesn't seem to be basic cultivation, so I figured I'd ask here.

Let's say I get a dried mushroom I like. Potency is good, everything is nice, and I want more. Is it possible to take a plug of the stem with a sterile syringe, and use to clone, or will a dried stem just not work for cloning? Thanks guys.

dry, wet, it will all work the same for cloning so long as there is still living tissue. there are many reports (ask RR) of dried-up mycelium being used to start new growth, so i suppose if you tried enough times, you could also use a dried fruitbody. people use gill fragments.

seems like hard work though.

I've successfully cloned from dried fruits many times. You want to first cut up a few pieces of stem into about half the size of a grain of rice, and then rinse them well in sterilized distilled water. Drop the stem pieces into the distilled water, put the lid on the jar and then shake like hell. This washes the stem parts. Then, transfer the pieces to another jar of sterilized, distilled water with a filter on the lid such as you'd use for growing grains. It takes about six weeks to two months, and you'll see a bit of mycelium begin to grow from some of the pieces. Transfer these to agar. You can use a syringe to selectively suck them out of the water. Don't use any nutrients in the water at all. Just PC'd distilled water.

If its dried and your a novice ... theres not much hope. It has to be fresh to get a good cloning. Revitalizing dried tissue is a more lengthy and iffy process. The one thing you could do is scrape the gills of the dried mushroom onto agar and issolate the clean parts.

If youve never worked agar you might as well start with something sterile and learn.

Is this gill scraping thing legit? RR? Seems like it would have to be hydrated somehow.

Scraping the gills is to obtain spores, not mycelium.

I dont see the difference in a drop of multispore solution on agar...and scraping the gills on agar with maybe a drop of sterile water...contamination factor?

You can try to rehydrate tiny pieces with distilled water, then clean them with some h2o2 before putting them on an agar slide. I wish you luck though because it's difficult to make a clean specimen out of dried fruits. You'd be better to go to the source and get a fresh one, slice it open with a sterile blade and cut an inside segment into a grain jar or something.
