12 is a good number. that's how many jars come in a box. and it's exactly 1cc per jar. that's enough to get some pretty quick colonization.

I did 6 jars and they were 100% colonized in about 2 weeks +.

depends on what strain. prolly about a week maximum, which i think twice the yield is definately worth an extra week.
