about old kit's spores
    #687058 - 06/18/02 10:51 PM (15 years, 8 months ago)

Hi. I just got a free kit thats said to be 6 yrs old. The spores in it are on paper in a bag which was opened 6 yrs ago. Can I still use these or should I just order some? Any info would be appreciated

They might be good....you might want to let the rehydrate for a little longer...maybe order some new ones just to be sure.

I think you should try them just to see. There isn't much data on spores that old. You would be doing everybody a favor by trying them out and reporting the results.

Actually, it's very easy to isolate a super producing sclerotia strain.

Follow the strain isolation technique on Let's Grow Mushrooms, and then select sectors early which are brown in color.  By the second or third transfer, you'll see stones developing, and this is only about 1 month after the original swipe of spores on agar.

Now, take each stone and move it to a new dish.  Soon, the mycelium will grow out and you'll see fresh stones developing, and if they're good strains, the sclerotia is forming long before the mycelium reaches the edge of the plate.  Pick strains which form four or five stones within two weeks and use these for your grain masters.

Yes it would be interesting. I've had a syringe 2 years old that innoculated fine. You might get results because its stored as a print. Let us know.

It says SPORE STREAKING, then has a paragraph about lysoling your table and burning alcohol swab to sterilize Inoculting Loop. Next paragragh says

"Remove the piece of paper containing spores and place it face up on the table. Open one of the petri plates, holdingthe top right over the medium to prevent contamination. Flame the inoculating loop, then cool it by placing it in one part of the medium on the plate. Scrape some spores off the paper with your loop and smear them in an "s" configuration onto the medium in the petri dish. Immediatly close the plate and proceed to the next one. When the flame dies on your alcohol swab, light another one. recommended to smear 2-4 plates"

Then you inoculate a compost mix after with small chunk from dish.

Could I put the spores in a needle?

Should I follow the instructions?

I wanted to try the pf-tek way, but I couldn't just scrape them in the jars.
I'm going to order a syringe and try the kit I guess.
