Liquid culture question
    #6834179 - 04/25/07 08:12 PM (17 years, 8 months ago)

So.... whats the difference between sticking some mycellium in some water with a stirrer...

or sticking some mycellium or spores in a liquid culture such as honey and having that stir....

The mycelium can eat the honey or whatever nutrient you put in there. It actually colonizes in the liquid. Mycelium isn't going to grow without some kind of food. Water alone just doesn't cut it if you're trying to grow a culture.

If you just want to create inoculant and not actually colonize a culture, then straight mycelium water is just fine.

But what is the advantage of colonize a culture?

You can put tissue from a single mushroom to clone it or create a ton of inoculant from just 1-2cc of spores. It just makes things a lot easier.

Guys who really know their shit like RR are going to want to start from spores on petri dishes and then make isolates from there. For people like me, the 'more casual hobbyist', multispore inoculations of liquid culture or cloning with tissue in liquid culture work well.

If you have mycelium, then you probably don't need to make more liquid culture, unless you want a bigger quantity.

For example, if you have 10cc of mycelium or spores, you can put that in a quart jar of nutrient-rich water and you will eventually have, say 400ml (instead of just 10cc, or 10ml) of mycelium.

The stirring breaks the mycelium up into smaller chunks so more gets pulled into a syringe, without clogging.
