Inoculated too much spore solution?
    #6810803 - 04/19/07 10:28 PM (17 years, 8 months ago)

My jars are colonizing right now and are at approx. 70%. I just noticed that one of my jars was contaminated and tossed it. I also noticed that in the other jars, at the exact spot I inoculated the solution, the mycelia was much weaker. The infected jar was also contamined right at the inoculation site. Might I have used too much solution in each jar? Would lowering the humidity level help ther survivors? Any tip appreciated!

If theres contamination in a jar during colonization, theres not a whole lot you can do about it. Humidity doesn't make any difference for jars, the jars are sealed. Unless you're talking about loosening the lids to allow some of the moisture inside to evaporate a little. Which I wouldn't do unless theres a lot of water droplets inside the jar. Just monitor the jars that you think might be contaminated and if they get worse, just bury them outside or something, who knows...maybe you'll go outside in a few weeks to find that there are...OMG, :mushroom2:shrooms:mushroom2: in your yard!!! lol, you can only hope;).

Next time just be more sterile in your technique and you shouldn't have contamination issues.

I got 3 jars with no contam... yet.  I am only worried because the inoculation site shows weaker mycelium than the rest of the substrate.

I've spread so much alcohol in my grow room, I was choking from the odor, hehe!
Hope those 3 cakes won't contam.

I'll throw the other one in my backyard, thanks for the suggestion, we'll see what happens!!

humidity matters as much in the jars as for fruiting substrates. standing water is always bad

No sign of contamination in my 3 other cakes, but they seem to have stalled at around 75% of colonization. What tickles me is that there is almost no mycelium at the inoculation site (where I could see the spore solution after inoculation).

To me, it makes me think that there was contamination right from the syringe.

I didn't flame the needle but swept it in alcohol. That must be where I failed, though I read some comments stating that alcohol flaming was not necessary. I guess I was mislead. Should've read more.

Maybe you're dry verm barrier somehow got wet during PCing or when you were inoculating.  Also, did you flip your jars?  You should flip when they're 40-50% colonized.  That really helps the mycelium take off and start colonizing the bottom.  Also, the reason those spots aren't germinating is probably because they may be too wet.

Thanks!

There is no way that swabbing with alcohol will remove contaminants that are inside the needle. Only flaming red hot will do that.

So I would be right to guess my other 3 jars will soon show signs of contamination (if not counting those uncolonized spots)?
