OK, well I put this in the advanced mycology, but realized it doesn't get replies fast enough. So since this is related to cultivation, I'll post it here. A mod can delete the other post if necessary. Anyways, here it is...

So, I got ahold of this really big cubensis strain, already dried. I used the cap and cut off some of the dark black veils for possibly spores that didnt drop. I transferred these veils to 7 different petri dishes with potato dextrose agar. I got 5 contaminated dishes and tossed those. I had 2 left and gave one to the original person i got the cap from. I saved the most rhizomorphically looking petri for duplication and looked like this:


I made a glovebox out of a comforter bag and 2 grocery bags and duct taped myself a ghetto glovebox. I sprayed it down with lysol and ozium to decontaminate it. I had 10 petri dishes ready. This time, I split the very rhizomorphic dish into tenths and distributed accordingly. After one week, it looks like this:


So, I'd like to ask you guys.

How do you think I'm doing?

Where should I go from here?

How long should I keep these dishes at room temperature until I need to do something with it?

Is it ok to put them in the fridge for long term storage without any ill-effects on the mycelium? And how long would it last in there?

What is the best substrate for this method to spread?

Should I consider liquid culture and inoculate that way?

Predicted date for use: June 2007

Do you perhaps have stairs in your house?

I will not answer your question my goon friend for I do not know the answer, but would someday like to obtain these skills, so I will be checking this thread.

I'd suggest taking some very tiny pieces no larger than a grain of rice from the leading edge of your growths and transfer them to new petri dishes to grow out. It usually takes several transfers to get ahead of the contaminants, and if you wait until they show up, it's too late. By taking from the leading edge of the growth, you're the farthest from the original transfer, and the least likely to have contaminants present.

It's good to see you get imaginative to make a ghetto glovebox to do agar work in. I take clear plastic trash bags to the forest to clone mushrooms in. We often find rare polypores such as fomitopsis officinalis and I don't want to have to pick it to clone. You can do work even in the field with the fruit still attached to the tree with those. Good luck with the cultures. Nice save.

I would also like to see the answers to your questions.

As I understand it, any kind of inoculation with colonized agar is fine, including LC.

I would put my money on bottom row, middle. Of course, you should do a few more transfers with smaller wedge sizes. Here's a transfer I made a while back from a dish that appeared to have uniform growth:
