first cloning attempt. using a variation of creamcorns cloning tek.

only difference being, i took the biopsy by push the needle up, rather than through the stem, in an attempt to avoid the contams on the outside of the stipe.

mistake #1. forgot to put some shrapnel,marble etc in the jar,so i couldn't break the myc up. mistake #2. no provision for GE, so decided to pc some fresh air and inject into the jar. obviously anytime you introduce a foreign object into a jar you risk contamination.
the "mother-ship" resisted all attempts to break her up,she seemed to have the consistantcy of a jellyfish. how do i extract a sample for the wbs i had in mind for her... then, a few days ago i noticed what looked like wispy smoke when i gave her a swirl, damn. did i contaminate her with my, probably unnecessary, fresh air injection.
anyway. i decided to inject her with 4ml of 9% h202 into her 500ml of "sugar water" in the hope of dealing with her possible contam. next day, her beautiful squidgyness had turned into something, resembling phleam oh well. i thought sod it,caution to the wind time. shook the buggery out of her and...she turned into this.

at least i could get a sample of her now. she was introduced to some newly made wbs. that was 6 days ago.

so, so far so good. she's a little slower than two other wbs jars inoculated with a m/spore l/culture on the same day. but,i'm guessing/hoping, her slow growth is due to the h202 i subjected her to,and that she will make a full recovery. we shall see...good vibes welcome

It is hard to get a good piece of mycelium with a standard needle and is best to split the cap and take a biopsy from the gore of the mushroom,do this in a glove box

yeah, i used a "sawn off" 18 gauge needle for the biopsy. wanted to avoid the cap all-together. didn't want to risk accidentally getting spores in the mix. this was done in open air.in the kitchen.
though, as a tilt at sterility. i washed hand/arms with bleach water and sprayed the area with it, donned a mask and otherwise practiced sterile techniques. must say though, i'd be surprised if this all succeeds.

Even though H2O2 will appear to "melt" mycelium in a liquid solution, it still does leave a good part of it alive, as long as the concentration isn't too strong.
Psilocybe Fanaticus had a tek on H2O2 cloning.
