Spore Printing/Syringe Questions(an idea)
    #6665206 - 03/13/07 05:35 PM (14 years, 3 months ago)

i was thinking about taking a jar that i usually use for cakes, sterilize and cool, pick a cap of a nice mushroom, place it in that jar, put the lid on the jar, place some coffe filters over the top, placing into my desiccant chamber until the spores fall and then...

...use clorox bleach wipes...place over the lid(after taking the ring-part off), slide the lid and filters out, use the clorox wipe to pick up the cap and remove while making sure to not leave any air holes(maybe by using a few of the clorox bleach wipes), then taking a sterilized syringe with water in it(boiled for an hour and givin 24 hours to cool), flame sterilizing the needle and alcohol swabbing it, then poke through the clorox bleach wipe(that's on the jar), squirting all the water into the jar, and then...

using the other sterilized syringes as the same as above but, of course, sucking the water with spores back into them.

the extra syringe would be to ensure that i get full syringes.

i wouldn't let the clorox wipes touch the water or print and no air would be able to get in unless it got sucked through the clorox wipes.

would this work? i dont want to make a glove box and i wont.

If you're going to do that, pretty sure a better idea is to turn the jar upside down and let the cap fall out, instead of reaching in there.

If your caps are not tiny & have thick stems/
sanitize a TREBLE fish hook.
Push it up through the stem.
So the EYE sticks out the top of the cap.
Then, place the cap in the jar.
When ready to remove the cap.
Grab the eye of the hook, with tweezers.
And lift the cap, straight up & out of the jar.

I plug the PF jar holes with silicone sealer and pressure cook with lid screwed on loose. I tighten it after the jar has come to room temp inside the cooker. I use a paper clip partially unfolded to pick up the cap and quickly place it in the jar. After it sits overnight, I use the same paper clip to remove the cap then let the print dry by leaving it partially open upside down over the edge of the lid. Then I screw on the lid. To make syringes I wipe the silicone with alcohol, heat the needle, cool it in alcohol and inject the sterilized distilled water forcefully to dislodge spores. I use 18 gauge needles that produce a strong stream. I let the spores soak overnight in the fridge before withdrawing the spore/water into the syringes. I wear surgical gloves and mask while performing these tasks, and I clean the kitchen counter thoroughly with a bleach solution, but otherwise I don't take extraordinary precautions. So far I've gotten many viable syringes free of contaminants.

another way to skin your cat.
sterilize a shot glass with alcohol
cut a hole in a piece of sterile card/tyvek whatever you like.
lay on top of glass
place cap on top of that and cover for 24 hours.
the spores fall through the hole in the card into your jar, without the cap ever touching the glass.
remove the card cap and all, and cover with another sterile piece of tyvek(allows spore to dry)secure with a rubber band.
finding ways to skin cats is such fun:eek:
