Is this the right Agar?
    #6609009 - 02/25/07 03:05 PM (17 years, 1 month ago)

If so...could someone please point me to the best Tek to prepare this stuff in order to bring back a dried specimen?

Thx.

That's the stuff. Bear in mind, it's just the jelling agent.

I've had the best luck bringing back dried fruits by placing in a jar of sterilized distilled water with zero nutrients added. Be sure to use a filter in the lid for fresh air. It will also help if you have a magnetic stirrer. It takes about six to eight weeks, and you'll see mycelium growth into the distilled water. At this time, transfer some of the mycelium to your agar mix to isolate away from any contaminants that may be growing along with it.

I use 20 grams agar agar, 20 grams malt dextrose, and 2 grams brewers yeast per liter of water.

Thanks RR,

What about a fresh specimen? how long does that take?
Or even spores that have gathered in my greenhouse?

BTW...I found this at a local grocery store in the Asian/Oriental Isle.
Don't ask the staff, cuz they'll just look at You like WTF and say no we don't have it.

Look your self for "Agar Agar" it's $1.35 per package.

Thx.

I've also read you can substitute table sugar for dextrose...is this true?

I will be using potato water also.

Thx.

Are you kidding me?!?! I paid $40 for my pre-mixed Agar/Malt after shipping. DOH! I've said it before and I'll keep on saying it. Ignorance and laziness are expensive.

Thanks guys,

I went shopping and being Sunday all brew stores were closed.
I have a couple questions I really need answered...

-Can I substitute dextrose with normal table sugar?
-Do I need brewers yeast or does normal bread yeast work?
-Can I use these...

...as petri dishes?

Thx a bunch.

To tell ya the truth, forty bucks isn't that bad for a pound of premixed agar. It's enough for about 500 petri dishes so it goes a long way. I've mixed it myself, and I've bought it premixed. I prefer the latter, but that's because I have too many other projects going, so it helps to let somebody else do part of the work.

When you go to the oriental food store, ask for 'auger auger' because that's how they say it. If you pronounce it aager, they will just look at you funny.

Ok...but I already have this stuff and was local.

Can I use the above things as petri dishes?

Those look a bit small to be of much use. I also doubt you could PC them. You want petri dishes that are about the diameter of your beer can.

I recommend the plastic, presterilized dishes. They're in a sleeve of 20. Don't open the sleeve until you're ready to pour the agar, and open it inside the glovebox. After pouring, put the sleeve back over the stacked dishes. It will help reduce condensation and will protect the unsealed dishes from contaminants as they cool.

Let your agar cool 'almost' to room temperature before pouring. I estimate my agar is about 100F to 110F when I pour. You should need no protection from the heat of the bottle for your pouring hand. This will cut down drastically on condensation on the lids.

It's cheapest to buy a box of 500 dishes at a time. It's amazing how fast you go through them once you get to isolating strains.

Well, guess I'll just STFU and use the search function...but it's just so vast when it comes to agar prep.

I have a very good idea...just a few missing links.
If anyone is kind enough to share there experience with agar...that would certainly be awesome.

But I'll tell ya...when I figure this shit out, I'm gonna write the best
Pictorial agar Tek ever!

Thx.

If you spend half as much effort on it as you did with your kick ass greenhouse tek it will really be an asset to many members of the shroomery including myself and I look forward to seeing it
