New Glove Box

"The thing is like 16 inches high, 20 inches long, 30 inches wide. I used a drill to make holes in the side and duct tape to secure gloves. The thing is practically air tight, but is a rubbermaid type container so the top has small gaps even when shut. I just spray the whole thing and contents down with lysol before I put my hands in the gloves and do whatever needs doing."

So, would you all have any tips for AFOAF? They opened LC jars to stir them with sterilized wire, took LC into syringes which were presterilized, and opened some PC'd WBS to inoc with LC. They'd only attempt opening the jars in a glove box, but they're hoping its not a big risk of contams. I appreciate any input.

I'd use a 1:10 bleach/water instead of that lysol to spray with, cheaper, and doesn't leave a residue that can cause mutations.
Try some foam weather stripping on the rim of the rubbermaid if you're concerned about it being airtight. Not sure how well it would adhere to the plastic, might have to sandpaper the rim, and rtv, or superglue it down, no ideas there, just a suggestion.

You don't need to have the box airtight. if roadkill can do g2g and work with agar in one of these then you can do all the work you need with your hands in the gloves and the box not sealing 100% Besides the fact that he doesn't have attached his lid consists of a piece of plexiglass set on top.

Oh yeah oust and alcohol are your best friends in this game. You should stop using that lysol for mycology purposes.

Thanks for the tip regarding lysol. I tried my best to keep it out of contact with the mediums, only using rubbing alcohol to sterilize tools which were to come in contact. Is bleach solution just as good as lysol for killing stuff in the air? Or should I just wipe, spray with 10% bleach, spray it in the air, shut, and then work? Thanks!

You'll be fine. I don't even have taped up gloves for mine. I just have a couple of 4" holes cut for my arms to stick through. I wear tyvek sleeves and latex gloves, but the thing is far from airtight. All you need is still air and you'll get that even with a loose lid. Of course, be sure to shut off all fans, central AC/heat, etc., an hour before doing any sterile work so the air in the general area can settle down. RR
