Tetracycline

Would the addition of tetracycline or minocycline to a typical substrate be beneficial to stopping contams or would it just slow mycelial growth? I know that anti-fungal drugs and antiotics have specific properties and that tetracycline especially wouldn't harm the mushrooms, but I don't know if I'd be able to heat either of these compounds, or whether I'd have to add them after sterilization, which would give more opportunities for contamination anyway. Does anyone know where I could get such information? (I have both left over as old prescriptions for colds and such....doctors prescibe them like water......it's a little irresponsible)

ive tried it and i did notice lower bacterial contam rates with the tetracycline that is

As far as I know tetracyclin is not heat resistent. Paul Stamets recommends gentamycin sulphate, which can be autoclaved. Fungi Perfecti used to sell malt agar which was premixed with gentamycin.

you can get tetracycline and many other antibiotics for very low prices at pet shops that sell fish. i recommend www.thatpetplace.com for mail order. also, it would be important to know what type of bacteria are causing the contamination at issue. most notably whether the contaminants are gram negative, gram positive, or if they come in both flavors that way you can select an antibiotic that will target the contamination. these antibiotics can be looked up online to see what they kill and to find other information on the drug. intuitively, it would seem that if one chose an antibiotic that only kills bacteria within a certain range (of similar types) one would be less likely to harm the mycelium. i used to be an expert on what antibiotics were of what type but it has been so long...

on the flip side, i do seem to recall kanamycin as being a very broad spectrum antibiotic. you could apply some kanamycin in water through a water mister on some mycelium and see if it kills it. if a broad spectrum antibiotic does not kill the mycelium then it would be great to use because it really will kill almost anything else. hmm, maybe i will try this. that is unless anyone has information indicating negative affects.

also, many antibiotics can not endure heat.

Be careful of any use of antibiotics, the effect of their use (read use = overuse in nearly all situations) is extremely detrimental in the long run.
If you only loose 10%, don't worry and perhaps better prepare your substrate.
If you are loosing more than 50%, you should reconsider your procedure.

I know that the overuse of antibiotics is dangerous in the big picture due to evolution of bacterial tolerance, but how can the use in a single casing be detrimental? I doubt the bacteria would recover sufficiently to pose any danger to the casing before it was spent anyway, please explain. Thanks!

in a single application, it will likely be effective at doing what you want.
But when, down the line, we are completely fucked because of this overuse.... you fill in the blank..
what is more worrysome about bacterial tolorance is that it can be shared by dissimmilar bacteria..
Staphococcus USED to be a benificial bacteria, and by in large it still is, but hospital-local infections are nearly allways due to staph, and it is increasingly resistant. There have been increasing numbers of cases of fatal infections.

I would not worry about contaminating your media with an antibiotic. Add it to the media after sterilization:

"...microbial contamination (of stock antibiotic solutions) is extremely rare, solutions that have not been sterilized are generally acceptable. If desired, however, solutions may be sterilized by membrane filtration."

Even scarier than the overuse of antibiotics is the fact that many bacteria are becoming more heat resistant (or rather,more heat resistant ones are being selected for) because of the food processing industry.

You know how it is often hard to sterilize rye?According to Stamets,45 min used to do it.Now he autoclaves for 2 hours!
