innoculating from Shiitake culture
    #641104 - 05/22/02 11:28 PM (21 years, 10 months ago)

Finally i've got my hands on a shiitake culture
I am in the process of making a glove box so thats one step out of the way, but i just wanted to double-check my interpretation of all the notes i've read.
To transfer the mycelium to the jar do you just use an innoculation loop or can you just use a sharp knife? Would it be worth making my own crude innoculation loop?
Once you've made the transfer do you have to do anything to the culture or do you just pop it back in the fridge?

Cut a square from it with a sharp sterilized knife and transfer it to a jar.
You will have to read some books if you want to keep the culture. It is not that easy.

why then do people bother with cultures?

or do you just have to do a large amount of jars at the same time.

Yes, I advise that you inoculate many jars at once, otherwise your culture will get contaminated.

I've read that its best to put the spawn into bags. Do you do this after you have colonized the jars? After the bags have colonised do you then case? what is the best mixture to case with? Or do you just pour the mixture into a tray.

You need cultures, specifically well maintained cultures to keep the strain you have healthy and productive. Typically grain spawn is inoculated into autoclavable bags of supplemented woodchips and dust. Shitakes are not a sure thing. You should really get a book or two. Oysters are better for a beginner in gourmets. MO.
