Queensland Albino on Kangaroo Poo

Ok, this has been a long time coming, but its been worth the wait!

Basically this log will consist of two grows that were conducted at the same time, one being an experiment using Kangaroo Poo as the substrate, with a strain I found about this time last year while I was in Queensland, in the North East of Australia.

While I was there I did a bit of hunting and found my first wild cubensis and an array of other shrooms which can be seen in THIS THREAD , including these crazy albino cubensis -



A stem sample was taken from the biggest specimen and stored in a sterile bottle of water, and a smaller sample was dried and stored for further study -



Upon my return home I began a liquid culture experiment using this Glucose -



- as the nutrient with the fresh stem sample, and thinking everything would would work out ok, threw out the rest of my fresh tissue sample.

That was a big mistake, as the glucose I used didnt work and I lost my fresh sample - the lesson - never experiment with rare samples!! :nonono:

I still had the dry specimen, and I thought it would be as easy as simply throwing a peice of that into a Liquid Culture and everything should work out ok - but after several attempts, I continued to get contaminated cultures.

I thought this strain may be lost forever and I realised if I was to have any chance of saving it, I was going to need the hand of someone more experienced.

I sent  a request to a certain 'Rabbit' that we all know so well, and he offered to have a crack at it. So, some samples of the dry stem tissue headed off on an exciting adventure, across seas and far away to the US of A....

A few weeks pass, suddenly came the news that they arrived safely and that initial experiments were underway... SEE THIS THREAD NB: These are NOT my photos -



A further few months pass, and suddenly the good news - after some initial hiccups - all was a success and the samples had been fruited, and prints were in the mail (now I need to stress here that this was probably one of the coolest things ever to happen to me within this hobby, and I couldnt wipe the smile off my face for a solid day after that...  :laugh: :laugh: :laugh:). NB: Again, these are NOT my photos -



In record time after the news, an envelope arrived from across the other side of the globe containing the spores of this interesting specimen. By now, he'd been on quite an adventure and covered more km's then most people I know!

I want to take this oppurtunity to thank Roger for his fantastic help with this project and stopping this sepcimen from being lost forever.

I decided at this point that it was time to utilise the interesting substrate which I had collected a few months previous to this to see if it worked.....

This is a kangaroo -



And this is roo poo -



It looked perfect to use as a substrate as the roos mainly eat grass and the poo has a straw like quality when dried and crumbled, just denser and more 'nutritious' looking than horse poo.

So, down to business... 

:mushroom2: Firstly, I threw some spores into a honey/water LC prepared the same way as in my signature and had a nice culture in about 14 days.

:mushroom2: I then inoculated 8 jars of WBS with 8cc each of the QLD Albino LC... in 6 days they looked like this -



:mushroom2: The roo poo was crumbled by hand until there were no chunks left, moistened to field capacity and stuffed into an oven bag -



:mushroom2: At the same time, some other substrate was prepared by soaking some straw, hpoo and coir overnight in a weak solution of seaweed extract, wrung out by hand to field capcity and stuffed into oven bags in a similar fashion to the roo poo, ensuring all the air was squeezed out of the bags. Then, all the poo bags went into a 'cooler' (we call them esky's In Australia) -





:mushroom2: Some water was brought to 180F on the stove and poured on top of the bags (they will float but this doesnt matter) -



:mushroom2: The lid was put on the esky, and its was left to sit for two hours. Just before going to bed, the water from the esky was poured back out into the pots, re-heated to 180F again and poured back into the esky which was this time wrapped in a blanket and left overnight to pasturise.

:mushroom2: The next day it was time to spawn. I made a mini terrarium for the kangaroo poo experiment by cutting a 3 Litre juice container, and covering the top of the bottle with micropore tape. The kangaroo poo was added and a small amount of spawn was mixed with the poo. I used only a small amount as I wanted to make sure it was actually the poo that was carrying any growth that occurred, and not the spawn that was used -



At the same time, the other bags of substrate were mixed with the rest of the spawn in two black plastic trays. A thin frosting layer was applied to the trays, and they were covered with foil sheets that had holes punched throughout and covered with micropore tape for gas exchange -



:mushroom2: All projects were put in a dark cupboard and monitored for any progress over the next few days. This is what the roo poo looked like on day three then again on day five -





It was fully colonised and looked very healthy -



So I decided to peak at the black trays and got a pleasant surprise -



:mushroom2: Time to case - prepared a mixture of 40/40/20 Verm/Peat/Coir with a sprinkle of garden lime, brought to field capacity and stuffed into an oven bag, then sterilised in the oven for an hour at 140C. I cased both projects about 2 cm deep, then covered the black trays with plastic wrap and poked a few holes for air exchange, while simply replacing the top of the bottle on the roo poo project -



:mushroom2: Back in the dark cupboard again to wait for the casing layer to colonise... About three or four days later there were spots of myc appearing on the casing surface so it was time to patch these babies, give a nice light misting and expose them to fruiting conditions - this meant covering the bottom of the orange juice container with foil to stop side pinning, but as you can see there are already knots forming so I was a bit late -





:mushroom2: The trays were put in a 90L plastic tub with a jar of water and a bubble stone running in, with a few small holes drilled and covered with micropore tape for extra FAE. I also supplemented FAE and humidity with the odd fanning and misting.

It ripped through the casing layer and continued to colonise a bit too much, temps were a bit warm (summer here and these werent kept in any special conditions) and maybe a tad not enough air exchange, but overall things were looking good for the trays -



:mushroom2: The roo-poo-juice-bottle was simply left at room temperature in the lounge room with natural light, and at night time lived on the fridge to keep temps up a little. This is what developed, as expected there was some side pinning -

 

 

Notice how pale the gills are, and the extremely thin, almost complete lack of a veil remnant -



:mushroom2: These were left to mature another day... Some of the fruits were completely albino, while the others were very pale -





A clone was taken of the completely albino fruit but is yet to be grown out. All in all, I think roo poo is a fantastic substrate!!

:mushroom2: Now this is the development on the trays in the fruiting chamber -





 



:mushroom2: I found about half of them were true albino's, and the rest either had a patchy spore print or just a very light spore print.  -



The later flushes also seem to throw off a few mutants, but Im not sure if this is because of the warmer temps they have been kept in or if it is a genetic thing (after the first flush I didnt care for them very well - not enough FAE etc. due to being too busy). This is a mushroom sprouting mushrooms (there were a few cases like this) -



:mushroom2: Finally, prints were taken and this very special strain can now live on through the ages -



This has been great fun and will be my last project for a little while - Im so happy this strain was able to be saved (thanks again RR for your help!).
