Scaffolding required in order to pin?
    #632278 - 05/15/02 07:30 PM (18 years, 3 months ago)

A friend of mine is try to do invitro jars with rye berries...he is now at the pinning stage, however, no pins are developing. In some jars that were innoculated at the same time using the PF BRF/verm cakes, pins are devleoping. My question is whether mycelial pinning requires the presence of some scafold material in order to pin, which would explain the lack of pins in the majority of the jars as they do not contain verm, however, some of teh jars had a light verm layer on top but no pins are developing either...

Could he add a light moist verm layer to the top and grow out of the jars from there? Or do these HAVE to be birthed into a terrarium setup and cased with something else in order to work? Thanks for any input.

i don't think they should require the verm as a "scafold" but it does tend to increase humidity at the substrate surface. the cakes might just be dry. have you checked for contams? if not, smell the jars sometimes bacteria can't be seen and can prevent pinning. if the cakes are just a little dry they might still pin but take a little longer.

Humidity at the surface may be a factor, however, as I have conveyed, my friend has had them under 4-12 hours light daily for the past 2.5 weeks and also in terms of humidity, the jars are definitely humid, the cakes may be slighty dry, but he can't seem to figure out other factors. The jars are definitely not cantaminated of this he is sure. Pinning has occurred with the jars that are BRF/verm even though they are not fully colonized, while the other jars that have been fully colonized for weeks haven't done anything.

He's in the process of dunking a a few of the jars, however, he feels that this is not the real problem. He'll try it out though. Thanks for the suggestion.
