I am assuming that it is just a trait of the species. It may be more problematic than useful if it tends to go into fruiting mode before a bulk substrate is fully colonized.

Workman have you ever worked with any fluorescent dyes on spores or mycelium? What types of stains would one use to show the internal structures of a spore?

Good luck with the new strain!

I don't know of any stain that will penetrate a spore or make the internal structure more visible. There are several dyes for enhancing the visibility of mycelium, even some common food dyes give reasonable results.

Another image has been added to the violacea paper.

Yeah it seems very problematic currently. Is the birdseed colonzing, or has it already fruited?!

Are there ways to make the strain not do this, by isolation or what? Seems like you're going the opposite direction in isolations. Rather than getting the strong fruiter, your first just trying to get the mycelium to colonize!

This is very interesting, I am new the the world of agar and strain isolation, I cant wait to see the process of how you're going to get this to fruit well.

keep up the great work, I look forward to checking out the results of your medium trials.
please keep us posted!

i was intrigued by the new antibiotic you were working with.
found some places that sell it, but i also found a few warnings about gentamycin sulfate being toxic, carcinogenic, and/or teratogenic.

what do you think workman? at 100mg/L it doesn't seem like much, but when it's come to cancer causing, and flipperbaby-makin stuff, i seem to prefer the longer road instead of the shortcut over a papermache bridge.
you know of any figures for this chem, such as the amount of safe exposure to it and so on, or do you suggest that I just go with my gut, stick to weeding out contams on peroxidated agar, and so on?
heh, are those warnings even true?

again, great work.

peace and thank you,
uniboner

I haven't looked into the toxicity of gentamycin sulfate. I assumed it was fairly safe when taken in theraputic doses and even safer when merely handled. I am fairly certain its not good for you in high doses over an extended time as with any antibiotic. I guess I should look into it and get a copy of the material safety sheet.

The violacea page has been updated a little bit.

I suspect they are not an active species of Psilocybe. No bluing at all. I will consider a bioassay in the future.

Very Nice bro!

great picture!

I love how they curl upward.
too bad they don't turn that violet color like the wild ones.
I was really hoping that these were active.
doesn't sound promising.

great work though!
thanks for sharing your research!!!

Well, some variation from the wild type is expected when grown under laboratory conditions. But I assume you are mostly referring to the violet coloration of the wild specimens. Upon closer examination of a larger, wider version of the wild photo, it seems that the purple color is merely from deposited spores. Spores from a mushroom that was removed before the photo was taken. This species is a very heavy spore depositor and the caps are pale and easily discolored by spores.

Errrr..... too late for that crop, but the next one is coming on from the same jar. They seem to be a bit more robust this time. The first flush was half developed in a sealed jar before I opened it and that may have had some effect on appearance. I really need to start over and make a cased tray.

There is risk. I think I'll try to send them to a lab first if they are agreeable. As far as I know, there are no deadly poison purple-black spored mushrooms. I could get very sick, but I would likely survive to tell the tale. I tasted one fresh and it tasted like bland mushrooms.

they look pretty similar to me.
Lab-grown specimens of most psilocybes tend to have a tendency toward more upturned caps due to higher/more consistent humidity.
I'm guessing that the wild caps are darker due to large spore deposits rather than any difference in cap colour, though even that wouldn't be surprising in lab conditions either.

The natural ones look to be coloured by pigment, not by spores. Is this the case?
Perhaps your sample was somehow contaminated?
The voracious fruiting on agar speaks somewhat otherwise, however, to contamination with other psilocybe spores from your lab..
Please do keep us up to date on your progress.

I think they look pretty similar. I would think it would be more amazing to grow out a contaminate that looks so similar to the intended target. I haven't previously grown anything like this before, so contamination from my other specimens is unlikely. I plated the original sample several times with the same results. The spores of the original sample match well with the cultivated specimen. It is a better match than the lab cultivated Psilocybe mexicana compared to its wild version as shown below.

I used a similar methodology to extract spores from dried specimens with both the mexicana and violacea.

The yellow mushrooms are Psilocybe mexicana next to a photo of wild Psilocybe mexicana. Check out http://www.sporeworks.com/gal8/8.html for more mexicana photos. And here for some general information about the species. http://www.sporeworks.com/mexicana.html

what you saw were enoki mushrooms grown in a high CO2 environment to promote long stems and small caps. If they looked like those in the picture here, they were harvisted too late.

I still don't see a pigmentation in lieu of the coloration from the spores.. it is very well possible that it is simply due to lab cultivation..
maby there is something missing in the substrate? some chem which is used to produce the coloration?
still wonderful work, regardless. The mini carpophores are definatly cool..
keep it up

Hey there,

Based on the size of the items that I saw in pictures in this article http://www.imaginaria.org/wasson/life.htm , the ones Eva Mendez handed to Wasson appear to be Mexicana also. The one thing they were not specific about was the exact item that were used in the ceremony that night...but I know some were sent on to Heim and I think Hofmann got his from Heim.

Best regards,

KingBolete

Overmature P. violacea in the same quart jar, third flush. New mushrooms developing on stems.

Now that is the most amazing thing I've ever seen in mushroom cultivation. I've seen caps with no stems, mushrooms growing upside down, a pinner growing from a cap, and plenty of joined mushrooms, but this takes the cake. This species has got to be the most determined fruiter I've ever seen, ever! Too bad its not active, it'd be a perfect cubensis alternate if it was active.

Anyways, great work WM! If only the mammillata would fruit so easily...:)

LK,

I wish you mucho luck workman Not to detract from this post or change subject, but have you noticed any sclerotia forming in the substrate or casing layers of the suspect mammillata? Just curious.

I failed again with the mammilata. I'll try something different this time.

Here is a picture of the violacea on millet cased with peat oystershell. Second flush started showing that pinning from the stems again.

The ring structure isn't the same and, most importantly, the spore color isn't the same. Identification on images alone is tentative at best.

Still amazes me that there are pins growing off the stems.

Has this species been proven to be active yet, or is that still unsure? Its considered a psilocybe, correct?

Also, how big are those mushrooms there?
