spore enumeration of suspension (real mycologists please)

greetings:

having trouble locating information on spore enumeration using a hemacytometer. I would like to count how many spores are in .5 mL of suspension. Do I pull out and Alequat,drop it into the hemacytometer, count it and then use this number to extrapolate to the .5 mL? Thank you very much.

If the spore solution is very dense, you will need to dilute your sample down so the spores are in countable numbers (around 50-100 per square). Shake the solution well to distribute the spores and carefully fill the hemacytometer well and cover with a cover slip. Each square (1 of 9 on the slide) holds a volume of 1 x 10^-4 ml = 0.0001 ml. So multiply your counts by 10,000 to get the number of spores per milliliter of your sample. You should do several squares and average them to get a more accurate count. Don't forget to factor in your dilution factor if you needed to dilute your sample. For example if you diluted your sample by 10, then you need to multiply your result by 10 to get the undiluted spore count per milliliter.
