Liquid Innoculation?
    #608415 - 04/14/02 08:18 PM (22 years, 1 month ago)

I am going to do a test with a few liquid substrate jars tonight. I am going to use organic honey for one and fructose granule for the other. Has anyone used fructose before? I belive a 1:10 ratio of honey:water. Any suggestions on the fructose:water ratio? I will probably use 1:10 as well. I will be using about 1cc in each 1/2 pint of a nice syringe of b+ that i just recived from The Little Guy. He was very fast with shipping and its nice and dark. Thanks to TLG

Dextrose & Dry Light Malt 50/50 ratio works great.

Teaspoon of each to about 3/4 quart water.

PC for 20 - 30 minutes. If it is cloudy or has residue (my FOAF always is), filter w/coffee filter & PC again. Looks about the color of light flat beer.

Same goes for honey/water.

Be sure to stack 3 or 4 peices of thick glass shards in the jars. If you get a batch growing well, it will get thick enough to plug an 18 or 19 gauge needle, when you try to suck it out. The shards are meant to cut the myc up, when you agitate the jar.

Drill a small hole in a metal lid. Use a small stainless steel screw to plug the hole. To draw from the jar, remove the screw. Draw & replace it.

If you use plastic lids, take several wraps around the jar threads w/teflon pipe thread tape. Plastic lids sometimes leak fluid when you agitate the jar.

Culture grows faster & better if you have some filtered air exchange.

Liquid culture is a magnet for mold spore contam's. When you draw from jar, make sure you use a sterile syringe. Otherwise -- contams. Hope you have sterile procedures down pat.

Hmmmmmmmm. Flat beer should work to, I wonder?

SixTango -- out

I've seen success with brown rice flour in distilled water. Just a teaspoon a so in a pint of water in a quart jar(better for swirling every other day or so). PC or boil. Some spores, and you're in business.

It has been about 11 days now and i have not seen much results out of any jar. I PC'd 2 jars for around 30 minutes. One with fructose 1:10 and one with honey 1:10. The honey jar has some stuff floating around in it, but i think it is just the honey byproduct. The fructose shows nothing. There are a lot of spores in the honey jar swirling around. I put 1/2 cc of B+ in each. I used pollyfill in the holes. Should i have a lot of growth by now? Any sugesstions to speed it up. ( it is in an incubator at around 82deg) I also tried doing a 3rd jar in the microwave and it has the same byproduct look. Can anyone show me some pics of what a liquid innoc jar should look like?

There are usually little white floaters in just a few days. By a week or two there should be floaters resembling tiny white jellyfish like you'd see at the city aquarium(size about 0.25 to 0.5inch diameter).

I've read several times not to add spores while the liquid is too hot. Also, make sure some air can get in--no vacuums.
