I harvested the large specimen today (see pictures in top post). It didn't seem to be getting any larger so I guessed that it must be mature. A good amount of spores are visible in the cut specimen as well as significant bluing. It smells potent and the flesh is tough like cartilage.

The "stem" appears very psilocybe-esque, woodlover psilocybe-esque. Would you say this is true?

looking good, what type of mycelium did this fungi produce? and what type of spawning substrate was used? you also said the fruiting temp was between 50-60f. what was the relative humidity? how long did the fruit body take to form? sorry about all the questions i find the cultivation of exotic species, very interesting.

Hi Workman Excellent presentation! Wouldn't that make a nice poster, I especially like the basidium shot. Just wondering if you had any more fruits form from your patch? Also do you have any micrographs of the Psilocybe arcana? I would be very interested in seeing them! Once again excellent job inski.

Thanks! The basidium was a bitch to find and I was only able to find the one. I am not sure why, since there were spores everywhere, but maybe they degrade after spore maturity. So far only three mushrooms have appeared from the patch. The mushroom in cross section is not the same as the one in the center of the picture.
I will do Psilocybe arcana next in a similar fashion. I plan to slowly do all of my collection this way. It will help me sort out subtle differences between strains and lock down some identifications.

Nice basidium picture. It's really hard to get those with a light microscope. I just ordered a new zoom microscope that goes to 200X so hopefully I can get some cool gill shots without having to make a slide, which destroys most of the basidia. Olympus claims it has a depth of field at maximum zoom of 20 microns, so hopefully it will be able to get the whole basidium/spores in focus.

the problem i find with basidia is that sometimes its hard to see whether your seeing a two-spored or four-spored one - and even when you alter the focus you can see whether the two spores you can clearly see are actually four or just two. In my experience - and im only just really getting back into the swing of things now - Basidia are easily the hardest to find - cheilocystidia the easiest, pleurocystidia pretty easy too providing you rehydrate well, and get a 'lucky section' (which isnt always just simply because its the thinnest), basidia the rarest generally. One interesting thing Im starting to notice is the changing cystidial shapes of specimens of the same species with age. Ive got to do a fair bit of more study on this but in some species the same collection can vary due to what appears to be the maturity of the specimen. Secondly, as Stamets and others have suggested, cystidia present in a species often do not correlate with the descriptions provided for species (Ps.cyanescens, Panaeolus cyanescens, Panaeolus subalt as examples). And given the huge variation found in diffrent collections of Panaeolus cyanescens in relation to cystidial shapes, sizes etc lets hope they arent as variable in other species or their role in ID is pretty much useless - certainly Guzman's reliance on them for specification seems of limited value.
