Ok guys, comments please on anything that could help/harm my project im a beginner at this so anything is appreciated. Ok heres my set up from start to finesh...
1 10cc syringe equador strain 10 1/2 pint jars
For each jar 1/2 cup Vermiculite, 1/4 cup BRF, 1/4 cup water, 1/2 inch of dry verm on top. Poked 3 holes in each lid and lids are on tightly, a 6x6 square inch piece of aluminum foil will be on top of that to keep out moisture from the sterilization process. Sterilized @ 15psi for 45 minutes in PC, Let cool for 24 hours.
Removing the jars from the PC i place them into a large rubbermiad container wiped down with alcohol for inoculation. Take aluminum foil off lid. Wearing latex gloves i remove my spore syringe from the package and sterilize the tip by applying a flame to the end of the needle and wipe off black residue with a clean washcloth dosed in alcohol. (sterilization is the key) Let the needle cool for a minute or two in open air and inject into the jars 1/2 inch below the substrate level, quickly i do all 10 jars. Put lid on rubbermaid container and using a heat pad i keep temperature inside the container at 82 degrees. (A thermometer is taped to side of container on the inside).
The Mycelia will grow
If im successfull in colonizing my cakes i will remove the jars from the rubbermaid container and set them aside. I add 1/2 - 1 inch thick layer of perlite to the container. (My incubation chamber has now become my fruiting chamber) I remove the cake from the jar, flip it over (latex gloves on) so the dry verm layer is on top and exposed and scrape that layer off, flip it over again so where the dry verm layer was is now on the lid, and place lid with cake onto perlite layer. Done for all 10 jars. Put on lid to rubbermaid container and lower to room temp, using a flourescent bulb on the ceiling i will provide the container a little bit of light this time.
(This will Induce Pinning and Fruiting in my cakes)
This is my project layed out pretty much but i have a few questions i need help with before i start...
1. How long should i let the needle cool after i sterilize it with the flame? Can i just wipe it down with alcohol?
2. Should i put .5cc in separate holes or inject all of it into one hole?
3. Should i provide ventilation to the box during incubation (leave a wide crack in the top of the lid) or have the lid completely sealed for incubation?
4. Do i have to sterilize the perlite? What kind of water can i use to dampen it and how much should i add per cup of perlite? Could i possibly use an alternative like clean wet paper towels/washcloth on the bottom to keep humidity up?
5. How often during the fruiting process should i let fresh air to my cakes? I was thinking Once or Twice a day of removing the lid for a few seconds should do. Can i get contams this way?
Thank you all!

Hopefully your needle is still sealed in the package it came in. When you get your needle from Ralphs, the cylinder is full of spores and the needle is still sealed. Don't do anything to the needle if this is the case. It is sterile as it will ever be. If your needle has been in the open. Take a little dish of alcohol, unscrew the needle and drop it in there.
Get a spray bottle and fill it with alcohol. Get a paper towel. Spray down your towel and spray the area you will be innoculating in. Spray your gloves, arms, etc. with lots o' alcohol. Wipe. When you quit gagging do it again. That headache from alcohol smell is the headache of sterility.
Try and get your jars fresh out of the pc when you inoculate. Don't let them sit around. Take your needle out of the alc and shake it off. Splash some of that on the needle cylinder. Let it dry a little, not a big deal. Now you have some sterile conditions.
Just squirt in one hole, move your needle aroud a little. Not a big deal.
Let air in the box for incubation. You don't need to keep jars sterile on the outside. Just spray them in alcohol before you dump out the spawn.
You always want air to get to mushrooms somehow. Filtered is better. Mold loves stale air.
Mix your perlite with water and peroxide. Don't be stingy with the peroxide. DI water is good.
Drill holes in your fruiting chamber and stuff with polyfill so they get air all the time. See posts with monotub setup.
My opinion - the key with sterility is to keep your time during inoculation short. Do a dry run by laying out all your equip with a used needle from somewhere. Practice it once so you have everything within easy arm's reach and you aren't knocking stuff around.
