First signs of colonization

Hello All, Hail to this FORUM! i'm a total newbie in shroom cultivation, though do have some knowledge in cultivating other herbs I have made 2 attempts to grow PC Golden Teacher with a PF rice-cake method. After first attepmt of inoculating the sterilized substrate in a jar, i stored it in dark place and have been observing it for a week. No signs of mycelium growth for 8 days! Not even a small white dot! Then i made a second attempt with all necessary precautions (i.e. jars are cool enough, water in the syringe is cool, etc.). The jars were enoculated with a newly prepared spore syringe, though i used the same spore print (got only one from a Dutch shop). now again, it's been already 4 days, and i see no signs of mycelium. I have undertaken a paranoid measures to sterilize everything around (i wiped everything with alcohol - working area, syringe, lighter, scalpel, rubber gloves, etc.). So, i assume that contamination might still have a chance, but i see no signs of it so far, no signs of mycelium either.. ) Can it be a merely due to dead spores, or i just have to relax and wait for another 1-2 weeks? Note that spores were brought from Holland by air, they were packed in a bag, which was sitting for 3-4 hours in a cargo-trunk of aircraft. Perhaps, there was a temperature drop to well below 0'C? Questions, questions..... Anybody ever experienced similar problems with the Golden Teacher strain? Thanks a million for your replies.. Partysan

What temps are you using to colonize the jars ? 4 days is a little early to see growth sometimes. some haven't seen any growth in over 14 days time. I usually see some growth in just a few days. Try making a incubator...and setting it at 83 to 84 degrees. If you haven't already. Good luck!

I've had spores seamingly 'ruined' by being in the frindge(or not being in the fridge..) I have no idea wat causes it tho.. I had a print that I successfuly spawned two different projects with.. I tried to use the same print about 1 month later......... NOTHING! think mother nature can be a bitch sometimes.. Peace

The jar s are kept in a dark place at roughly 75' F - room temperature. I monitor the temp with a thermometre..

the print was in a ziplock baggie, inside a Grower's kit, which was delivered from Amsterdam. The kit has been stored at room temperature. Is it better to keep the spores in a fridge?

yea but they will still rehydrate in the jars if he didnt wait so this wouldnt be a cause for too much worry..

Aloha 2 all farmers! Hey-hey-hey, i checked the jars today morning and guess what, the micelium is there... Hehehe.... 5 out of 7 jars have bright white spots. Interesting note - that old jar, which didn;t succeed last time, still shows no signs.
Last time I filled it the verm+brown rice powder, which was included in a Grower's kit from Holland. I re-sterilized it and tired this time, but still nothing.. This is a tall 250 ml glass, previously filled with French mustard. Of course i washed it thoroughly, but maybe the mustard smell, which is hard to kill, is a problem for fungus? Or i assume that Dutch verm-rice is not good. Note 2: one of the jars had about twice the amount of water in substrate mix, than recommended by PF tek. This jar has the most robust looking micelium.
