How To : LC Flasking DIY Glassware.
Liquid Cultures are best accomplished with a stir plate. However many hobbyists make do without. My method to increase yeast and spawn cultures uses readily available materials at little or no cost, using common liquor "Hip Flask" bottles.
Materials :
Half pint / pint bottles ( 200 / 375 ml ) with plastic caps.
Self healing ports or,
RTV for DIY ports.

A step drill bit is very handy for making clean, accurate holes in the cap.

First simply make a hole in the cap by drilling from the inside using a scrap of wood as a backer. The backer helps prevent cracking the cap if too much pressure is applied. Be sure to make the hole a snug but not tight fit for your Self healing plugs. That’s all there is to it. The flasking cap is ready to go.
If using RTV make a hole up to a half inch in the cap. Wipe cap down with alcohol to clean. Using a cotton swab ( Q-Tip ) apply a little petroleum jelly or thick soap water around the inside of the cap, covering lower thread area and sealing lips. You don’t want RTV stuck where the bottle neck goes.
Place a goodly dollop of RTV on soaped flat surface, described in RTV technique linked above. It should be about as round as the cap. Run a ring of RTV on caps top, around the hole then place cap down on the dollop. The idea is to smooch the ring lightly into the dollop, making a single glob of RTV that protrudes through the cap.
Let RTV cure then clean away petroleum jelly, if used, and boil to fully cure RTV.
Your flask caps are now ready.
Fill your DIY "Flasks" half full of liquid media. Use a funnel or wipe bottle neck with alcohol to keep clean. Cap bottle and unscrew about one eighth turn. Cover cap and Flask shoulders in foil and PC as usual. When cool tighten cap till inoculation time comes. The large volume of air in the Flask creates quite a vacuum. You may inoculate through foil if you wish.
After inoculation, the Flasks may be shaken for effective aeration and breaking strands. Simply grasp at neck and vibrate side to side, sloshing culture. Do this several times a day if possible to keep myc broken down into smaller bits. If allowed to float and mat it becomes challenging to break into bits for use.
CAUTION : Do not use marble agitators as in jar culture. They are not really needed for this method and the Flask will break, making a real mess.
To withdraw LC for inoculation, use an air port. If you don’t have any air ports you may sterilize syringes with the plunger pulled back. Be sure to have a half cc or so of water in the barrel to make steam during PCing. When working just push a cc or two of air into the Flask and withdraw a corresponding amount of LC. Several reps will have your syringe full for use.
