Psilocybe cubensis "Un Id" isolate on straw/compost cased.

This is Un Id colonised rye. I isolated fruit bodies off of a multispore agar plate from what apeared to be a healthy strain from what I will call the Un Id strain. This jar was inoculated with an agar transfer on 3/28. This is my first rye jar that did not develope a bacterial contam. I am contemplating on what to do with it. I have considered spawning straw w/ it or possibly some more compost, maybe my tastey mint compost.
This strain feels good to me. I have noticed strong growth from the original multispore through all of my isolations. I hope to be able to distribute spore prints from it soon.

Transfered fruits from previous plate to new plate. This is after 7 days:
I dont have a final pic from the previous plate once it grew out, but I can asure you it was a beautiful site.
Here is the colonised rye grain after 10 days:
All in all, three isolations were done from the original multispore. BTW..this is my first attempt at fruiting from an agar isolation.

looks like a piece of tyvek under the lid there, is that it? I?d like to know since I have an everlasting supply of tyvek postal mailing bags. I?d like to know how well they work in actual use.

No, its not Tyvek per se. It is similar though. I got the filter from a trade. Anno has had great luck w/ Tyvek. Use it by placing over the mouth of the jar, place lid and then ring over the top of that. If you need further explaination let me know.

I decided on spawning this w/ a modified Dr. Bluethumbs compost. I took his compost and mixed it with straw, vermiculite, and spawn mate from his site. I sterilised this in the oven at 350 F for 1 hr in an oven bag. container is a diaper wipes container. Substrate is a little over two inches deep.

Yes, please do dazedsol. foaf considering some outdoor cultivation in the future. Do you know whereabouts in Fl these were found? I?m sure I?d have better luck growing outdoors if I started with a strain that already likes my area.

Hey yea......you guys know about as much as i do on these........they came from a friend down in Fort Myers.....thats all i know.....i have been trying to get ahold of my buddy down there for a while now......but to no avail......i?ll let you guys know if i find anything else out......they do seem to grow quiete large though....some of the prints i had of these were huge, the size of a minidisc.......

I had a set back. The casing material I added previously was contaminated. I have eliminated the contamination. This is what the compost looked like before I cased it a second time just now.

I am using the floor of a closet as my grow space. Unfortunately the floor is a wonderful place to find contams. I also beleive I have a high mold/spore count in my home. I am working on being more careful w/ my practices. This casing that contamed was from the mix itxelf. I pre-sterilise my casing mix. I had this one in a quart jar covered w/ foil. It turns out that I puctured a hole in the foil w/o noticing before cased my substrate w/ it. As soon as I can I will be moving my set-up to a better location. I have been keeping my eye open for cabinettes that would work. This summer I will have much more time on my hands since I will be on vacation from school. You should see some nice projects then.

You could use a large rubbermaid to raise it off the floor.
Stick your growing chamber on top of the extra container.
Plus you could put some of your supplies in that unused container.
Like the ones that I use in my grow room.

They are not growing upwards. I thought my lid allowed enough diffuse light, but by the look at things I may need to improvise.
I also added additional casing to the tray. It may look like I took the second picture closer, but I scaled them both to be the same size.

You are witnessing what I believe to be the first captive growing of this strain. The fruits are from a multispore isolated fruit off of agar...I hope that last part made sense.
A close-up of the larger fruit. This cap is just past prime pickin?s. If I would have caught it about 1/2 an hour ago it would have been perfect for a bioassay. I am choosing to print my fungi friend. Check back, I will post at which point to harvest the cap.
4 hrs later it is starting to drop its spores.
This is another small shroom developing a little behind the other two.
4 hrs later.
I know they?re all out of proportion. I am trying to leave them in the terrarium and not move them so much due to past contam problems. To get the angles right is a bit hard.
If anyone has software available that would allow me to post moving .gif? images I would appreciate it. Road, I?ll PM you about this, I think you said you might have the program I need.

You called it as you saw it...I see no problem in that.
I think I isolated part of my problem. I have been using a styrofoam cooler as my fruiting chamber. I think that the porous surface of the styrofoam harbored contaminants. I have since started using more H2O2 in my cultivation and have switched to a Rubbermaid. I will beat this and have wonderful fruits to show.

Yeah thats a good point actually. The styrofoam could have really harboured some contams in the structure of the terr. actually. Willamette is pretty damp and probably a good area for contams to flourish.
Your fruits are looking cool. Cant really compare them to anything cause you?ve never seen ?em before, but thats a sic thought..

Are you going to clone the big one? If not, why? Im trying to learn about cloning, and the right situations in doing so.
If you cloned the big one, theoretticly, couldn?t you get a dense flush next time? And then you wouldn?t have to deal with spores...?

I am majoring in Biochemistry/Biophysics, I hope I explain this correctly.
The fruits you see are from a fruit that grew from a multispore isolate on agar. Since it was from a fruit it is diploid, meaning it has two sets of genetic material. The fruit you see has the same set of genetic material. By cloning the fruit I would not accomplish anything since I already have an isolate of it growing on agar. What would be more beneficial would be to take the print from the clone which would be haploid (one set of genes) and grow it out either alone or with another print from the same strain. This would allow the different sets of haploid mycelia to mate creating a different dipliod fruit. I could then clone that fruit and work from there. By repeated selection I would eventually get a gene pool rich in desired traits.
Ultimately creating a good strain through cloning requires several sets of srong genetic material. You would want to take prints from the strongest fruits and mate them together. By cloning a fruit you are bottlenecking the gene pool. You are only allowing the more desireable genes to propogate. In my opinion one can not rely on clones to grow consistantly, maybe for a generation or two. Eventually the strain will start to senesce? sp. Senesce refers to the degradation of a strain due to being grown too long on the same substrate. I have no idea whether the last couple of sentences is true. I base my opinion on the fact that a healthy population usually has genetic diversity. What I intend to do is create a strain that has good diversity but also has been selected for desireable growing characteristics.
So all in all, cloning and genetics is too deep of a topic for me to cover. My advice to you is:
Select strong fruits and clone them if you want consistant and coordinated flushes. If you want continued strength and diversity in your fruits, mix genes by using multispore prints from different clones.
Again, this info is from my limited knowledge, please take it as so. It would be great if Anno could offer some of his advice and expertise on the subject also.

I?ve purchased the TMC a while ago, read everything they said about cloning, and I have somewhat of a grasp on what to do. They dont go into the gene pool topic too much, if not any. But basically, I was thinking if I could clone the biggest fruit from my multispore flush, and create some spawn with the agar. Do this, get the fruits, and if I tried cloning the biggest fruit from that, and did it again, would that work. Hm I need to read more I guess. I can?t find too many good links on this however.
Im just trying to figure out the methods of preparing agar now.
Would you mind explaining your agar preperation?

If you clone a clone you get the same clone as you started with. This would not be advised as it would be a waste of time.
Buying a genetics text book might help you to understand. I find genetics to be very interesting as it is easy to see the effects of in our everyday lives.

Weight of second flush was 89.9 grams wet.
I cased the tray again w/ contaminated casing. This is a pic. I got green mold as well as cob web mold. I pushed casing into the gaps in the sides of the tray. Needless to say I had contamination all over. I am currently treating the tray to rid the contamination. As soon as I am sure the contamination has been neutralised I will break the substrate up and recase. You all should see pictures w/i the next week of my healed casing.
The white spots you see would have matured to nastey green mold, the cob web mold is pretty aparent.
