Since you can not put spores onto peroxide agar, could you make a small grain jar, knock that up and use a single colonized grain on your plate for making an isolated strand?

i dont use h2o2 tho after i am done putting my wbs grain on the dish i press it down lightly and then spray the top of the petri with lysol shake it then put the lid on and wrap it in cling wrap never had a contam yet :-)

What is an isolated strand? If you meant to say isolated strain, that isn't how it's done. You want to isolate many strains so you can fruit them independently to find the best performer. That means starting from spores, then isolating the various zones of separate growth that appear on the petri dish away from each other, so that each can grow out on its own dish. Don't use peroxide. Instead, use sterile procedure. RR

i think what he is talking about is starting a multispore grain on a petri then doing some strain isolation trasfers till he gets an isolated strain. then from there he is going to see if it is a fruiting strain and make an lc out of it. right
but he just wantd to know if he could use a piece of grain to start his dish..instead of spores.

The reason being is that on peroxide agar you can not put spores, only live tissue or such.
And I am not 2, just not the best speller that is all

Is there pure agar agar out there? The agar mix usually contain sugar in the mix. Is that alright to use for isolating strains in petri dishes?
