GROW LINK: http://www.shroomery.org/forums/showflat.php/Cat/0/Number/5934922/an/0/page/0

1) Substrate is mixed, with Vermiculite, brown rice flour, and calcium carbonate first. Add water after 3 dry ingredients are mixed then add water till it has fine subtrate texture. Spray a lil Hydrogen Peroxoide/Water solution. 1 part H2O2 to 10 parts water is what I use.
2) Use clean XS ziploc containers and add the subtrate to about 2/3 full from the top. Do not close lid. I filled 12 of these.
3) I put 3 of these containers in the microwave in a triangle shape on the microwave turning tray. I microwaved for 2 minutes, took them out, closed the lid and continued on with the 9 other containers each 3 in a microwave at a time and closed lids after
4) I waited 10 minutes and quickly opened lids to allow equalization of pressure and sprayed the top of the subtrate with 2 squirts of the 1 part 3% H2O2 to 10 parts water. I then quickly closed the lids, waited another 10 minutes, and repeated step #3.
5) I repeated step #3 and step #4 for a total of 3 times
6) And then on the last one, I used an x-acto knife to cut 5 holes on the lid like so. I used the x-acto knife to make pizza shaped cuts and then used the pliers to fold that pizza shaped cut back into the lid, to leave a hole for inoculation.

7) I microwaved all 12 containers one last time for only 1 minute and after put the tyvek filter ontop, then the vent filter. I put the foil ontop of that and only folded down and waited 2 hours to cool down completely to allow inoculation. Here is a pic of what it looks like:

8) After the containers were completely cooled down, I inoculated 12 containers with three 12 cc syringes. That's a total of 36 cc for 12 containers which is 3cc each container. That is 3/5cc per hole btw.
9) After inoculation, I quickly recovered the lid with the tyvek, then the vent filter, then the foil which i rubberbanded like so:
note: this picture has 2 used syringes for those 8 containers and one syringe which is the "secondary" syringe. The main syringe are those dark syringes which are master syringes.
10) After all containers were inoculated and filtered, I put them in a dark box with a centigrade thermometer and kept it in around 25C-35C.
11) It has been 9 days and now have 90% colonized jars with ONE contamination.

 contamination sucks...STAY CLEAN!
IMPORTANT INFORMATION TO GROW THIS TEK: I believe the reason why the microwave has a high failure rate is because the people that use microwaves arent necessarily that cautious of other factors that can lead to contamination. First of all, I don't use solely the microwave. I use Hydrogen Peroxide and sprayed my substrate with it since the microwave evaporates alot of the moisture. I spray the ziploc substrate in between microwaving. Also, I spray twice.
I quickly open the lid and spray. Since the ziploc containers are not hard, they will shrink and the sides of the container will shrivle up. That is why it is necessary to leave lid closed, and open it every 10-20 minutes to allow the pressure to equalize, and so the container will keep its shape. Also, I spray 2 times everytime i open the substrate container. I make sure NOT TO SOAK the subtrate, and to keep the subtrate as porous as possible. I don't want it compacted so the mycelium can spread faster.
Also, you must thoroughly clean all working areas and the microwave to ensure sterilility. Clean the microwave by hand and then after it looks spotless, use rubbing alcohol to wipe the microwave down. You can spray disinfectant in there. Make sure to microwave for 1 minute to keep microwave free of the cleaning supplies. I also wipe down my kitchen with rubbing alcohol.
note: I never used a glove box, I just spray everywhere with disinfectant and wipe down workign area with rubbing alcohol. I wipe all my instruments with rubbing alcohol and also use a torch lighter to kill all contams on the needle.
Just be sure to remember STERILE!!!

Agar did something along these lines, months back. http://www.shroomery.org/forums/showflat.php/Number/5397786#Post5397786
From what I recall, he could get the plastic containers to fully colonize, and fruit. But, after repeated attempts. He abandoned the testing, because the results were erratic, probably due to a multitude of variables involved with microwaves.

do you not think it a little preliminary to declare this 'foolproof' when you've already lost 1 of 12 and the others aren't even done yet?

DAY TWELVE: Dunked in 1 part Coffee, 89 parts filtered water, and 10 parts 3% H2O2. Going to be in the fridge for 24 hours then put into fruiting chamber

DAY THIRTEEN:
Put 8 cakes into fruiting chamber, after dunking for 18 hours in 1 part coffee, 89 parts filtered water, and 10 parts 3% H2O2.

why oh why did you dunk in water with coffee and h202. also you shouldnt be dunking in the fridge, cubes are a tropical species and dont respond to cold shocking.
Wouldnt it have been much easier and more reliable to just use a big pot of water to sterilise your jars??
Seems like your using a mix of all the old outdated and terrible teks, escept with using the plastic containers.
Use a glovebox for doing inoculations.

I realize this is an old post but I am a newbie and I think I just might try this but what about wiping with alcohol instead of hydrogen peroxide or using both
