So I had 6 premade jars sitting around...

I ordered a bunch of jars when I was a complete noob over a month ago from some company, premade substrate sterilized substrate jars. Anyway, they've been sitting above my sink for weeks now. Today in the mail I received some syringes (Should be the last ones since I started my first LC tonight :smile: ) - anyway, I inoculated 12 jars regularly, then I remembered about those 6 jars sitting up there.

They were sealed with lids so I didn't know how to go about inoculating without exposing to contams, this is the procedure I used, let me know if you think this will work or not;


Cleaned off kitchen table, sprayed air down with disinfectant, kept a bottle of hand sanitizer handy. Got 4 small nails with a hammer, sterilized each nail beforehand with lysol wipes and a paper towel soaked in alcohol, sterilized the tops of the lids. Hammered the nails down about 1/4 inch into the jars and left them there on all sides. When all 4 nails were still in the jar, tore off a piece of aluminum foil, sterilized both sides with alcohol/lysol wipes, pulled the nails out and wrapped up with alum. I did all of them like that, then cleaned out the bathroom real good and inoculated regularly in the bathroom.

Think these jars will come out? Can sitting there that long ruin their sterilization? But if there weren't sterile, wouldn't a contam start growing before I even injected the spores? I was very careful to re sterilized everything in between jars, my hands, the nail, the jar, and double did it with lysol wipes and alcohol. Also when I punched the first nail in each jar you could hear the air slowly slowly escaping so I think it helped keep any airborns out.

Anyway, that's what this post is for, feedback. They're being incubated at ~83 now in my first incubator, just curious as to whether this was a decent procedure or not. The only sketchy part to me seems like the foil but I made sure it was pretty clean.

Sounds like you did everything you could have done.

Only thing I can think of is that there was most likely a vacuum in those jars if they were totally sealed, and when you punctured those holes, weather you heard it or not, some unfiltered air was allowed into the jars. That air you heard was air rushing into, not out of, the jars.

You dont have any filter on the jars now as of what I read in your post, so I would get something on there like a coffee filter, micropore tape, something.....and cross your fingers

Oh, and for clarification they've been in the cabinet for about 2 weeks now, or so. Still in the package they came in, too.

Right not they're sitting in the incubator with the lid (4 holes punched) and a piece of aluminum over top of that. I'm going to leave it tight for a few days then loosen the aluminum and perhaps replace it with coffee filter. Is that sufficient for now?
