does anybody know exactly how many spores are needed to be loaded into a syringe? all the tek seem to be very vague on this topic.

its all opinion on that subject. you only need 2 microscopic spores to mate and you get germ but they are microscopic so you just cant count them.lol .i usually load enough to see some little floaters in there other people use a whole print. some just use just a little. i guess it is just preference.

HAHAHAHA
Sorry that had to be done.
In a glove box with a clean scalpel scrape off some spores into a clean shot glass of distilled water and suck it up with a clean syringe.
If you want to get technical you need 2 spores to properly germinate.... Unless you want to look under a microscope and pick them out just do what i said it do.
This is why it is called multi-spore injection

as many as you like. some people use a whole print in one syringe. i would sometimes call this a waste but i also do it.
normally i use about 1/8th of the print just to make sure their is enough but not wasting the print. useing more spores gets you a slightly faster colonization but creates many substrains from the amount of spores all forming their own network. they meet up and join together as one with no problems... hope this helped a little. ~thenewguy

As a follow up you are better off makeing an LC. Then when you get more advanced make a master cloned LC.
LC is liquid culture and can basically turn a few spores into possible gallons of spore solution

it does not have to be done in a glove box if you dont have one. it is better to but i have never used one to inoculate and i have had 3 con tam out of two hundred jars or so. just make sure you spray the air, close of all drafts, sanitize the counter plus the stuff your working with and wash your hands good.also wipe your needle with alcohol and flame your needle tip you should be fine.

i put exactly 847,234 spores in each syringe.
hahah no i make as many as 10 from a single print, sometimes as few as 1-2. depends on how dark and how big the print is and how many empty syringes i got kickin around. you could do even more than 10 with a nice big print. that's really why its so vauge, everyone does it a little different. its really hard to go wrong, even when your syringe comes out crystal clear theres still many many thousands of spores in it and it should do the trick. if its tinged the slightest bit purple when you h old it to a piece of paper, you got more than enough and it will work juuust fine

Like Babo said, an LC is deffinately the way to go in this case. I was amazed when I tried going from print--->LC. I scraped a really really small portion of it into some sterilized honey water, left town for like 4 days and BAM I had a big blob of thick mycelium in there (which, with time, got even bigger). And with only one small liquid culture you can make so many syringes which will work better (quicker) than multi spore inoculation. Give it a try man!
