i have some rye grain in 1/2 pint jars im ready to case. can i make a 3/4 inch hole in a lid, tape, and pc my grain. then when im ready to case transfer a little of the rye substrate into the hole of the sterilized jar and cover hole back up with tape. i read before of someone doing this but adding a hydo perx solution to the jar and shaking fist. any ideas of if thats needed. if so what about misting the substrate im gonna put in the new jar with hydro peroxide. what ratio of water to peroxide 3%.

Give the colonized rye a one minute bath in pure (3%) peroxide before transfering to your fresh jars.

Well, personially, i would put a polyfil filter floss filter in it, and depending on your inoculation method, go from there
if you'r egoing to xfer from a colonized grain into the new batch (spawn) i would just have the one hole /w polyfill. if you're going from spores.. maybe a second hole covered with tape would work. not sure how you inoculate through a polyfil filter floss hole.. maybe someone who uses spores can tell you.
you may be able to stick the needle down in beside the filter.. depending on how tight it is.
as far as the h2o2 coating the rye after PC, i dont know how necessary that is. but if you're going to be doing it in an unsterile area (not in a flowhood) maybe it would be a good idea.
ive never seen a problem without coating in h2o2.

when I first tried g-g transfers I would soak the colonised grain in h2o2 and transfer to peroxidated grain, and I'd alwaysget shitloads of wetspot.
Now I just shake up the master jar, remove the lids from the jars, pour a little colonised grain into the freshly sterilised grain and replace the lids. No flowhood/oven tek or anything and 100% success rate.
I think the h2o2 slows the mycelial growth so that any contams can get a grip before colonisation, whereas fresh unperoxidated grain colonises so quickly that any contams don't get a chance.

I think you are right about h2o2 somewhat slowing the growth. i have came to the same conclusion.
