Best simple method I have found is to use a large bore (14g) needle. (on a sterile syringe) And, take a core sample. (PREFERABLY - working in a glove box)
First cut stem about 2 inch in length. Drop in hydrogen peroxide for a few minutes. Remove stem, insert needle into stem. While needle is inserted, bend stem, rotate needle 360 degrees. Withdraw needle. Needle should have a core sample in it.
Inject core sample into a prepared LC jar.

It depends on your wants & skills.
Cloning will often get you good genetics.
Core sample clone will get you the best of several worlds. It's quick, clean & effective. A decent LC can inoculate a barrel of grain/wbs spawn.
