Some quick questions guys (and gals). First, does anybody know where you can find a alcohol lamp? And if i cant find one is the flame on a gas stove ok to heat up needle? How long should you wait after needle is red hot to inject?

you can just use a lighter.
and you get it red hot, then squirt a little spore solution out to cool it down, then you wipe the needle with alcohol to clean it off

Science and surplus store.. or ebay. Andy thing is fine to heat it up. Just as long as it is red hot.. U can use a butane torch lighter if you want. The benefits to using the alcohol lamp is that u can have a stand to hold the syringe.. (best for agar since u need to have 8 hands) I am cheap.. I just use a lighter.

After what he just went through i would have to agree its not a good idea... just make sure the alcohol evaporates !!! Safety is number one. If you aren't there the shrooms wont grow!!!

not really. it's a very simple thing to do. I put less than a 1/4 of an ounce of alcohol in the glass. It's really not as dangerous as it sounds.

Here is what I do to make a very nice alcohol lamp. I am always experimenting with fire and so this lamp that I am suggesting is pretty safe if you're not foolish with it.
Materials: ~Rubbing Alcohol ~Shotglass (or a similar small, heat-safe cup (I use the cut-off bottom of a soda can) ~Cotton balls ~Aluminum foil
Fill the shotglass half-way with alcohol, and cover the whole opening of the shotglass with the foil. Punch a small hole in the center of the foil to have the hole leading into the center of the shotglass. Unravel a cotton ball and get it soaked in alcohol. Weave the alcohol-soaked cotton-wick into the shotglass through the hole in the foil.
Make sure there is NO alcohol on your hands, and light the cotton wick.

Adjust the length of the wick sticking-out of the foil hole to adjust the length of the flame.

You can find an Alcohol Lamp Construction Kit (TM) in your own kitchen.
1- take a little jam jar (empty) 2- poke or drill a hole in the center of the lid 3- (optional) poke a little airhole in the lid 4- stick some cotton wool through the big hole to make a wick 5- fill with alcohol 6- screw lid on jar 7- light
(You only need the little hole from step 3 if you have drilled a perfect hole for the wick, without it the air pressure in the jar starts to rise as the lid heats up, causing the flame to grow very large, the airhole makes for a more stable flame)

Nice writeups Mushouse and koppie, but if you plan to use your alcohol lamp more than once I would advise getting a real one. Saving $3 isn't worth working with a homemade alcohol burner.

So if you you heat up the needle do you still need to wipe it with alcohol? Or can you get away without doing this?

No, you don't need to wipe it. Heating anything red hot completly sterilizes it. I always thought it was kind of dangerous to wipe a red hot object off with a flamable substance, but then again I've never heard of anyone being burned from doing it.

after that needle cools down and is no longer red: it is again available to be contaminated.
within a glove box with still air, or in front of a laminar flow hood, you will probably be okay with just the flame sterilization.
in any open air sort of inoculation, flame sterilization is not enough. like I said as soon as the needle cools, it can contaminate.
the hot needle should be cooled in an isopropyl swab and contained within the swab the entire time. never touching open air.
if the needle touches open air, it should be flame sterilized again to ensure the inside of the needle is not contaminated.

So heat up red, close off in alcohol swab, then inject. I dont have a glove box so i will just heat red, close off with alcohol wipe, move to pan with hood over stove sucking air up (and a couple burners on to keep hot air moving up), inject then re-heat.

I don't use a flame , do my inoculations in open air , and never have any problems just wiping with alcohol .

It's not advised to use alcohol to wipe a needle that has been flame sterilied. Simply heat the needle red hot, then use. Don't wait for it to cool completely down. The first drop or two of solution will cool the needle, allowing the rest to flow uncontaminated. RR

I DON'T flame anything , never have in 2 years . Maybe you missed my point ,lol.
From my understanding , most the oldtimers don't either anymore , that's who told me not to deal with flaming needles anymore . And I haven't had any contamination issues .
When I started I made the alcohol lamp from the baby food jar and cotton . Then bought an alcohol lamp . Then threw them all away after nearly burning the house down. Flame sterilization is nothing but a PITA if you ask me.
A wipe with alcohol seems to work for me

I had doen flame to needle at first (last year). I have had terrible trials this year with just alcohol. I might try both but i feel better seeing the red hot needle knowing that isnt anything in the air around me that could survive that.
