malt/dextrose sediment in LC with stir plate

If one is using a stir plate would it be beneficial to leave the sediment in the LC (from the malt/dextrose) since it will never be allowed to actually collect on the bottom? I would assume the sediment would just add to the nutrition of the LC.

I dont like it, but dont know if it harmfull or not,

Though it does look like contams, that's way I go to length to filter it, with this there is no sediment.

i never remove the sediment, its fine, but that usually means that you are using too much malt/dext. i use 1tsp of each per pint.



also, stirplates arent kept on for 24 hours a day typically, they can be but it is unnessiccary.

Table spoon each light dry malt & dry dextrose, into a quart of boiling water & then filtered through 4 or 5 coffee filters into a 1/2 G jar, then PC'ed @ 15 psi 20 minutes. Day 5 today.

I've done my 1st malt/dextrose LC. Nothing grew. So I will try again.

Ive read that the idea amount was 1/2 tablespoon each per cup of water. Or 1 each per pint, which is more 1 each per quart.

on my last LC run, i went ahead and ran a few tests to figure out the proper amount.
this is using light malt extract and dextrose without filtering:

16oz of water in a Quart jar.

Malt / Dext Results
1 Tbsp each - Way too thick, slow colonization, excess sediment
2 tsp each - Good colonization, some sediment
1 tsp each - Colonization equal to 2tsp, but no sediment.
1/2 tsp each - Healthy, but less thick, colonization.

i use 1 teaspoon of each in about 16oz of water.
i see no need to filter with this amount. i get strong, sediment free LCs without wasting time filtering.

i changed it slightly, try 1 LEVEL tsp / 16oz h2o.

i forgot that my LCs are actually closer to 19 ounces, so i use a heaping tsp.

i got the instructions from mycofile.

1 level teaspoon of each too 500-700ml of h20 in a qt jar.

3% is pretty standard. 2-4% works well. All these measurements are just confusing. Just remember 2-4%. That holds for karo, honey, malt, whatever.

For people adding dextrose to their malt media... Have you found that to be helpful?

Malt Extract Agar (3%)(MEA)(General Microbiology)(FDA M93)
----------------------------------------------------------
30 g Malt extract
20 g Agar
1 L Distilled water

Boil to dissolve ingredients. Avoid overheating, which causes softening of agar and darkening of medium color. Autoclave 15 min at 121°C. Dispense 20-25 ml into sterile 15 x 100 mm petri dishes. Final pH, 5.5 ± 0.2.

This medium is recommended as a general maintenance medium.

yes... i find i like dextrose much better than adding

Malt + Karo  or
Malt + Honey.

i also have sediment,  but give it a good swirl on the plate about once or twice a day and you'll be fine. I just made 4 different strains on 6/29 - MS inoculation.

Two of which are looking real good. :wink:
