can't break up LC

I've been shaking daily for a while now, this PESA liquid culture.

There's a marble inside the half pint jar as an agitator.

No matter how hard I shake it I can't break up the mycelium enough to suck into a syringe.

Any advice?

yep...rock the jar back and forth until you get small enough pieces to suck into the syringe. you might not get a lot on the attempts because it has become so tuff but keep trying and you'll get a little.

I never use an agitator; I just suck the myc up as much as possible into the syringe and purge it back into the karo water a couple of times to break it up.

This last time I spent a good 3 minutes trying to suck a clump of myc into the syringe, but it just wouldn't go.

There was a huge vacuum created in the syringe since I had pulled the plunger all the way back (so there was like "negative air" in the syringe) and no myc or water would get into the syringe.  Then I released the vacuum slowly and noticed that there was about 1/4 of a cc of water in the syringe and as I was purging it back into the water, my plan was then to just start franticly sucking up water and shoot it back out at the myc in an attempt to break it up.

The thinking behind this is like that game that I used to (and I admit that I still do) play in the bathroom:  I would throw a few squares of toilet paper into the toilet, whip out my wang, and try to tear the toilet paper up with my quick powerful blast/stream of urine...

Anyways, I purged that 1/4 cc of water and the now broken up myc shot through the karo water, creating nice wispy strands throughout it like usually happens.  Then all was well. :syringe: :thumbup:

try a larger sized needle. i know most wal-marts will sell needels without a script. or try online

While I realize this doesn't help you at all this time, try a magnetic stirrer next time around. They're dirt cheap to make, the LC colonizes much faster and the myc is broken up into easy-to-aspirate bits.

Arrakis is right,That is what I had to do just the other day,I had a mexi-cub LC that was one giant clump.I shook it and shook it and only some of it would break up.It clogged the syringe bad so I just kept pushing the plunger up and down a million time into the LC till it finally sucked up now the jars are knocked up and the rest is looking great all broke up in a bunch of vacutainers ready for the next time I wanna take a look at mexi-cub.Good luck man

I'm actually having another liquid culture problem.

I've got a Golden Teacher LC that grew successfully, and the marble seems to agitate it very well.

Turns the whole solution milky sort of when I break up the mycelium at the bottom of the jar with the marble.

Well, I used all of a 10cc syringe innoculating 4 jars to test it out. Nothing. It's been over a week.

What could I be doing wrong? The jars don't have any signs of growth of contams or mycelium. Just wet spots where I injected the LC.

What kind of LC is it?
Karo, Honey or other type?

What kind of substrate did you inoculate?
How much LC did you inoculate with?

you can get larger syringes here as well as all the 18G needles you can shake a stick at. A larger syringe would let you creat more vaccum and most likely allow you to suck up just about anything. You can also get sealed vials which could be helpfull. As well as rubber seals to create the perfect re-usable lids.

(I know they aren't a shroomery sponsor but they are helpfull and no sponsors sell this type of stuff)

I'll definitely check out the larger needles.

I used Karo Light and I injected about .5cc into each of the 4 ports of BRF cakes just as I would inoculate using spores.

I'm looking at the syringe and I can't really see any mycelium in it. I'm thinking I really just might not have sucked anything significant up. Then again, I did use an 18G syringe and those pieces of mycelium were pretty darn small after I shook it up.

I can't speak for all needles but the connectors on my spore syringes are the same as the connectors on my flavor injector, cullinary syringe.

There are three common types of syringe barrel tips. Luer-lock, slip tip, and cath(catheter) tip.

Luer-lock and slip tips are interchangeable. With slip tips the needle just slips on. With Luer-locks you twist it on and the "wings" of the needle twist into the threaded sleeve and keep it in place. It doesn't matter which one you use. If you plan to put a lot of pressure on the plunger to spray out the contents then use a Luer-lock so that the needle doesn't get shot off the end. Otherwise slip tips work fine unless you plan to really pound that plunger in hard.

For the Luer-lock to work you need a Luer-lock barrel and needle. If either one isn't a Luer-lock then it will just work like a slip tip. Most all needles are Luer-lock, but they don't always say it. They are interchangeable (at least all the ones I've seen) so don't' worry about it. Just avoid the cath tips, they don't take needles but catheters instead.

bump ~ I hear that if you use a lot of sugar in your lc, when it hits your substrate if the sugar content in that is significantly lower than what it's used to it might not take to that substrate.
