> Each time you turn on that magnetic stirrer or shake that jar of LC. You are creating another generation of spawn/innocculent.

Why would you say that? Cell division isn't going to be changed all that much by stirring. Colonizing a LC is going to give you a P value of around 3-5 I would guess. It won't matter if it's shaken or stirred. Colonizing the LC is still going to require the same number of cell divisions, it'll just happen a bit faster.

I don't agree with you. By your reasoning if you take a cake and cut it in half you are now on the second generation. By that reasoning if you cut the halfs in two you will then be on your third generation and so on. That doesn't make any sense. By cutting something up or breaking up the myc you haven't really done anything to it. It's still the same myc with the same number of cell divisions.

A generation is a completion of the sexual cycle, not the number of cell divisions. The parents are the first generation, their offspring is the 2nd generation, their offspring is the 3rd and so on. What you are talking about is the P-value, which is the number of cell divisions required to colonize a petri dish. P-value doesn't equate very well with LC, but the principle is still the same.

I actually think the opposite of what you're saying is true. Imagine if instead of one piece of myc in the center of a dish you put 4 pieces. The dish would colonize in one quarter of the time and each piece of myc would only have 1/4 of the number of cell divisions that they would have with only one piece.

Myc cells divide by adding on to the end, so the ends have the greatest number of cell divisions while the center cells have very few. If you break up the myc then it will start to divide from each of the NEW ends which will be younger than the old ends, so the entire culture will colonize quicker and be younger than if you just let the natural ends divide to fill up the space.

So I say it's probably BETTER as far as delaying senescence to stir the culture and break up the myc.
