Using spawn bags /?

while reading about these spawn bags i purchased, it said to inoculate with 140 ccs of liquid culture..aproxiamately how many spore prints would that consist of..??? and also what is the best substrate to use?? right now ive prepared 4 bags with Black Cow,wild bird seed,ryeberries,vermiculite....if anyone knows any better recipes for better or quicker results,please let me know..

You are correct Jstump20, 140cc per bag. You need to have a liquid culture ready to go first. 1cc of spore solution will knock up 4 coffee cups of liquid culture nicely. That will give you about 450cc of liquid culture solution for inoculating spawn bags.
Here is what I do:
Using a full coffee pot of water (12 cups), I use 10 grams of dextrose and 10 grams of light malt extract for every 4 coffee cups of water. I am specifically saying "coffee cups" because a coffee cup is less than 8 oz. or a standard cup. Okay, so that equates to 30 grams of dextrose and 30 grams of light malt extract for every full pot.
Place the dex/malt into the area where you load the coffee using 2 coffee filters. Run the machine. This will yield 12 cups of liquid culture medium. Load these over 3 quart jars, cover and PC for 25 minutes at 15 PSI. Allow to cool overnight.
Inoculate each jar with 1cc of spore solution. Agitate the solution everyday with either a magnetic stirrer (stir bars loaded prior to PC'ing) or with marbles prior to PC'ing. You should have healthy colonization and be ready to inoculate within 7-10 days. Your solution should like something like this when it's ready:

From there, follow Blue Helix's Spawn Bag tek.
Here is a substrate recipe that I use and works very well.
Substrate Mix for Spawn Bags
Yield: 4 large filter patch spawn bags
Ingredients: 48 Cups Shredded Horse Manure 24 Cups Vermiculite 16 ½ Cups Wild Bird Seed 4 Tablespoons Kelp Meal 6 Tablespoons Canola Oil 21 Cups Water
Directions: 1. Mix all dry ingredients together thoroughly. 2. Add water gradually and mix by hand. 3. Add Canola Oil. 4. Load bags evenly. 5. Pressure cook for 2 ½ hours @ 15 PSI. 6. Allow to cool for one hour and remove from cooker. 7. Impulse seal bags on setting 5 for 5 seconds. 8. Hang to cool.
I typically have full colonization between 8-10 days. Each batch yields between 8-10 ounces dry, per flush. At least in my experience.

CARLITO, i have some quick questions about this...after quart jars(light malt/dextrose/1 cc spore) are colonized,you then pull the solution into a 140cc needle right?and then knock your bags up..if so how can you get solution out several times and still keep the solution in the jar sterile..??Do you have to take the lid off each time you need more solution for other bags?
and also after i have a few quart jars colonized with the solution,can i store them in the fridge for later use??

Correct. You draw the solution into your 140cc/ml syringe.

By using either a vacutainer injection port, which is what I do, and swabbing the needle with alcohol every time you inject. The only real chance for a contaminate is in between injections. The quart jar and spawn bag should both be sterile.

Again, I use a vacutainer injection port. If you have to do an open air aspiration, I would highly recommend a glove box. Here is a pic of what I'm talking about.

I routinely keep my liquid culture jars in the fridge for months on end. They can be stored long-term for later spawning to another liquid medium or grains.

You get them from a vacutainer. Vacutainers also make great storage vehicles for spore and liquid culture solution and can extend storage capacity for several months or even years.
You can either get these on the net or shoot me a PM and I can probably get you some.

How do you know when the solution is fully colonized and ready to be drawn into a needle?? and when origanally knocking these quart jars do you use the standard method( pop a hole big enough for a needle to get through,inject 1 cc,then put a coffe filter over it)?

Your first question was already answered with the posted photo of a colonized LC. On the second question, you could pop a hole and stuff it with poly fill for inoculation. Simply put your needle through the poly fill and draw out what you need. Worst case scenario you'll need to replace the poly fill if it gets solution on it. This is not the recommended course however.

why is there such a large hole on the vacutainer port lid wronguy? You also might try to find some 14 ga needles when shooting up with thick LC. I just ordered a full box from the farm supply for 40$ for 100. They did not carry 1 1/2 needles. That should last me damn near forever.
Also I am still spawning to pastuerized mixes. I like it until I can get a cleaner process down. How full should you fill filter batch bags and when in the pc how do you keep them from taking on extra water. Like jars you cover with foil. My bags do no look really wet but they have a lot of moisture in them, but they are not slimy or soggy at the bottom. PC time 2 1/2 hrs
