Hmm...
I dont kno why.. but I seem to constantly get contam when I do a spawn transfer... I have used the oven tek... this last time I went as sterile as possible...
in oven
coated with lysol
latex gloves
open one jar, cut cake with clean alcohol coated knife
quick dump into filter patch jar
seal
Proll 1.5 seconds of open time on the jar
Still got contam.
any ideas why this might have happened... or even better what to do next time...
would soaking the cakes in an H202 mix prior help... or spraying it... how can I safely transfer spawn!?!?!?!

Oven tek doesn? t work, use a glow box or make or get yourself a flow hood.

Hmm....
I was thining about a glove box.... I am probably missing something... but how do u make it sterile inside the glove box....
even if u can sterilize it .... how do you introduce items sterily? Does this work? where is the good glove box tek?

I used to have the same problems as you with contamination. Ditch the oven tek and make a simple carboard/saran wrap glove box and your problem will most likely go away assuming you are using good sterile technique everywhere else.
Anno had a picture posted not too long ago showing a jar on a hot oven rack with a piece of burning incense beside it. The airflow from the hot oven actually pulls the smoke INTO the jar. I verified his results using bubbles and found that the only thing the oven tek is good for is keeping particles well above the work surface away. Anything below the work surface (90% of all contaminates live within 1 ft from the ground) get stirred up as you work and sucked right into the jars. IMHO the oven tek is junk, though lots and lots of people on here swear by it.

why are people saying the oven tek doesnt work? it sure has worked wonders for my friends.... id still be wiling to say it works better than open air, yes ive seen the smoke... but the smoke came from down low, i dont think the contaminants would actually make it down near the jars with the hot air rising.... just my opinion... and i know afoaf that always uses it and never gets contams

I'm not saying to switch to another method if it is currently working for you, but if you are having problems and don't know why, perhaps the oven is your culprit. I had a kitchen full of 1cm bubbles and every time I moved, or the fridge kicked on, the bubbles would fly around with many of them landing on my work surface. For my oven in my kitchen the oven tek does not work. I had contamination problems similiar to the poster, and when I quit using the oven tek my problems went away. -shrug-

Ya thanks a lot for the advise... and the assurance that I am not a stupid person. I just found out today that my sch00l has a clean transfer system and an autoclave in the BIO lab that I can have full access to. Mwhahahahahha... Too great! I guess my problems are solved.. Hope they dont think its odd that I am autoclaving 50 quarts... hahaha.

I spray a bunch of lysol in there about an hour before I am ready to use it. If you are using a flame, remember lysol will burn, so be careful.
