Well I did a GTG transfer about a week and a half ago. Here is what I did I washed out a rubber made tub with atibacterial soap. Then I rinsed real real good. Then I rubbed with alchol and cut holes. THen I put gloves on and put through holes and robbed alchol on them. Then I took a knife dipped in alchol for a wile and then used it to push the fully colonised grain into the fresh grains.
Fresh grains history.
About 2 weeks ago I pced them. I left (maybe my error?) The tinfoil on them and took it off befor the g2g transfer when I took it off there were some puddles of water on the tyvek.
Well about 1 1/2 weeks later little green spots showed up.

It seems to me that you already see where you made your errors live and learn my friend. The only thing i see is that you should concentrate on construction of glove box FIRST and THEN worry about disinfection right before use

I meant I made the hole a little wider My arms didnt fit through very good it only took 5 seconds. I doubt that got contams in. Do you think the knife coulda done it since It was only diped in alchol? Does alchol kill spores? Should I of pced it first?

70% isopropanol alcohol works great to sterilize tools and surfaces. It's the only thing I use besides soap and water.
There is usually a bit of moisture on my tyvek under the tin foil as well and it doesn't cause any problems. Pulling your arms in and out of the box can cause suction that will pull in outside air. Perhaps your glove box wasn't sealed up fully. Did you add any additional seal to the lid of the box?

no additional seal I thought it was based on still air that makes it contam free how do I get the air steryl?

A still air glove box should be fully sealed. I like to just put micro pore tape around the lid.
I believe the idea of the still air box is that any contam spores will settle at the bottom of the box. They greatly reduce the chance of contams with G2G jars but you still need to do other sterile practices like doing it a reasonably clean room and having a clean flat fabric free surface to place your box on. You should also do the transfer as quickly as possible.

I say your glovebox was fine. It doesn't have to be hermetically sealed. A still air environment is what you want, and yours would take care of that fine.
A puddle on the tyvek in a grain jar is an almost guarantee of contamination. Under the foil, the condensation would be coming from the grains. A wet filter with nutes is a perfect path for contaminants to colonize right through the interior of the filter and into your grains.
The second mistake was using the knife to scoop out your grains. Alcohol is not a sterilizer. You need to break up the grains by banging the jar agaist a fully inflated tire half a dozen times. Each kernel will separate from each other kernel that way, allowing you to pour from one jar to the other without ever touching the grains with a foreign object. RR

When I do G2G I just set up in front of a flowhood and use two fresh plastic spoons that I keep in a glass of isopropyl. What I'll do is open both jars, take the two spoons out of the iso cup, scoop out a little mycelium, and push it down into the grain in the jars, drop the spoons back in the cup, replace lids and move on to next jar. So far this has been successful.

I've always done my g2g transfers in open air. I use a isopropyl-saturated towel to clean an ordinary metal spoon and simply scoop a little bit from a fully colonized jar to a PC'ed one. In all the times I've done this, I've only had one contamed jar ever.

that's what i do soulsizzle. i've only done 4 g2g jars, but they're all perfect, so yeah.
