agar plates

I just purchased a package (20) pre-poured PDA plates. The instructions that came with it said to allow the plate(s) to warm to room temperature before use and keep in the dark. I've set one aside for use for later, but, seeing as how this is a new adventure for me, I'm a little nervous about the looseness of the lid.

By simply moving the plate into another room and putting it in an old wooden chest of drawers, will any air enter the sides of the dish and contaminate my plate before use? Would it be wise to wrap the warming plate in parafilm, then unwrap for use, then re-wrap?

I am not too familiar with that but yes you want to wrap parafilm.

I know to wrap them in parafilm afterwards, but I'm wondering if I should do it before using the plates while they warm up to room temperature. Thanks, though.

After you put your multi-spore / cutting use parafilm like you said. I then pit them in a plastic zip-lock bag then into the incubator. Or you could use a ruber maid sandwhich container, both work :smile:

Never use a warming plate with petri dishes. Leave them in the bag they came in until they are IN your glovebox or in front of a flow hood. Wrap with parafilm, four squares per dish as soon as you inoculate. There is absolutely no reason to worry about keeping petri dishes in the dark. Light has zero effect on agar or colonizing mycelium.

thank you for an answer. I wasn't using a warming plate, though. I did however, pull one out of the sleeve and moved it somewhere else, but now I will open it and let it contaminate, just for fun.

I guess I'll set the sleeve out to let it warm up and then start.
