Liquid Culture Strain Isolation

Two mushrooms were found, one very young and one very old. Prints were taken but nothing dropped out, at least nothing that could be seen with the naked eye.
Scissors were used to cut out a circular shape where the caps used to be. The circular shape was then cut into quarters to produce four strips of paper. A liquid culture made from honey was obtained and the strips of paper were dropped into the jar.
Four days later an extensive mycelium network could be seen clearly residing on the bottom portion of the jar, along with the strips of paper and a single human hair. The was shaken regularly to aid in distributing myceclium and nutrients around the jar.
A syringe was used to transfer mycelium from this jar into a plastic container under sterile conditions. The second container is now full of mycelium.
Can the method of using portions of previous liquid cultures making new liquid cultures eventually produce an isolated strain that is contaminant free?

No. Any yeast or bacterial contam. will be transferred every time to the new culture. The only way to isolate is on agar.
Sounds like you're worried about the hair? Maybe it was in the media before you PCd it. If so, it's fine. Otherwise your culture would surely be contaminated and will probably become cloudy.

Also, don't confuse strain isolation with isolation away from contaminants. They're two totally different things. However, neither can be done in lc. As said, you need the two dimensional flat plane of a petri dish. RR
