Help Finding LC Jar Tek
    #5645634 - 05/18/06 09:18 AM (17 years, 8 months ago)

I'm trying to find this tek I saw once, my search attempts have turned up bubkiss. Help!

I think this tek was by Agar and it involved jars with tubing (automotive windshield wiper fluid line) stuck in it, polyfill in those tubes.

Anyone know?

Its all  in here

Although I find a simple 1/2 pint jar, with 1 small hole (slightly larger than the needle so you don't have pressure problems when drawing liquid) poked in the lid, covered with a piece of electrical tape works just fine and is so simple we could train helper monkies to mind our LCs.   Gas exchange really isn't a problem with LC in the first place, it gets a little bit each time a syringe is drawn from it.  Can be shaken to hell and back and the electrical tape keeps it sealed.  To each their own... I just like things simple as possible.

I think I'm getting my teks all mixed up.

I want to stop using micropore tape because when I PC the jars, the micropore becomes pretty much welded to the lid. I cut up my hands something terrible last time trying to clean it off. So I was thinking two holes in my jars. One small RTV'd nail hole for the syringe and a polyfilled tube for gas exchange on the other end.

Thoughts?

micropore doesn't fare well in the pc for me either.

the tubed lid is for liquid culture only.

for grains,

2 holes...like you suggested
1 rtv injection port....like you suggested

1 hole is for gas exchange...
no tube with polyfil added

you could use just poly-fil in the hole, but then no need for the rtv port.

many experienced growers put only one hole in their grain jars.
stuff this one hole with poly-fil.
inject right through the poly-fil. No problems.
don't worry about contams wicking in.... don't worry about it.

if you don't want to use poly-fil or micropore, you're down to my favorite filter:

tyvek. can be placed above or below the lid, screwed down tight with the band. Easy and effective.

whatever you do...place 2 coffee filters over the top with a rubber band for a dust cover and foil the top of your jars for pc'ing to keep the moisture correct.

thats why i sterilize with no tape on the hole but tightly wrapped in foil... quickly remove foil, inoculate, and stick a piece of tape on right away after.  i use electrical tape because then its totally sealed.  again IME i'm not concerned the slightest with gas exchange in a LC and never had a problem, but i know others will argue til they're blue in the face that you need all these fancy setups to provide it... i think a half filled jar with a big air pocket in the top is just fine.  with micropore on an LC, if liquid touches the micropore, it kind of seeps through and you end up with drops on top, and basically a wick for contams to travel right through the tape and potentially into the jar... not so good.  with the electrical tape it stays shut, nothing can seep through the tape, and you can shake the crap out of the whole jar.

oh, liquid cuture.

I can be quite an idiot sometimes...

yeah, creamcorn has great points.
agar's lids work very well also.

if you don't like either and want better gas exchange, check blue helix's lids out. I currently use them, but have the same turn out as with agar's.

agar's are quite spill proof and easy to aspirate from.
blue helix's are not spill proof and are harder to draw from.
creamcorn's idea sounds good to me. Gas exchange doesn't seem to be such a huge deal in my experiece either.

man, all these different lids every where.

My first lc wasnt doing anything till I put micropore tape over the hole for air exchange but we all have our different set ups. try a few and find one that will work. I put my micropore tape on after I pc now, much easier to work with. I dony have much room at the top of my jars either and that might be why i have to exchange gas more then others(just a thought)
