So far, I've tried nearly all the plastic wraps, micropore medical tape and masking tape. None can compare with parafilm. You want gas exchange, but no air exchange. Micropore tape gives too much exchange and the dishes dry out. The Saran type wraps either don't allow enough gas exchange, or none at all, resulting in slow growth. Masking tape doesn't stop contaminants. I'd get parafilm. It only costs a few pennies per dish to wrap and does the job well.

Use cotton or pollyfill to plug your tubes. You can take a roll of saran wrap and cut it into rolls about the size of packing tape. The main reason to wrap is to seal out contams and prevent the medium from drying out too quickly. Parafilm is a lot better like RR said, but it is kind of pricey.

Try inoculating two side by side dishes with the polyethylene on one and parafilm on the other. What I found was both cultures would grow pretty evenly for the first week, then the polyethylene one would slow down, while the parafilm wrapped dish kept growing rapidly. Neither contaminated however, so the Glad does its job that way.

I would see this as an advantage, you have to plate less often, or?
