silly question....

If i take a single colonized seed from a WBS jar.... can i place that on a agar petri dish for cloning?

using some tissue from a mushroom would be considered cloning.

you can do either...they both are going to grow mycelium.

using tissue from a mushroom garuntees you got a strain on your hands that will fruit... there's a chance that you get unlucky with the random seed you pick out

got a point....


also i just recently bought some vendor syringes...

i put a drop on a few plates, and i now have trich insides, with a tiny white myc outer ring??!? any reason why the drops contamed?

If it is not airborne contamination or poor medium sterilizing the syringe must be contaminated :runaway:. Anyway you could transfer the clean mycelium to another plate :sunny:

NO - if that jar has been started with a spore syringe.
as that would simply bomb your agar with various substrains.

yes and no, it will grow mycelium yes.

but basically the same mycelium that would grow if you simply started spores on agar.
anyhow, you could grow out the plate and start selecting and transferring various substrains originating
from the kernel.

Right. There will possibly or likely be many sub strains.

The white outer ring probably isn't mycelium, it's probably mold that hasn't quite sporulated yet, wait a day, I bet it will turn green.

Don't be so impatient, wait for fruiting, and pick your best fruit, then clone that. You'll be guaranteed of a great fruiting sub strain and in the end you will have saved time and most importantly peace of mind.

The grain will work but is almost a waste of time in my opinion, personally I suggest you clone the best performing fruit.

unless he wants to try at isolating strains on agar,
but then he could already start by streaking the spores onto the dish
and skip that grain part.

yes the contaminated ones are trich for sure... (discarded)

the grain i just wanted to try and see if it will work, and it does... i have had issues with Ms innoculations, but thought that the agar wedges grew very rapidly.

even havign a substrain on agar is a-ok with me. I will develope them and see what fruits. As for Cloning, i have ready petri's for that Big-un that pokes his little head out i will clone that and will def. post pics.

Edit: what's with streaking the plates? i just droped a drop of vendor bought syringe on the plate... that contamed. how would i streak it with a vendor bought syringe?

this actually should not be the case as it could indicate that the sporewater is contammed.

are you sure that the contams just gew where u put the drop of spore solution,
if so, that syringe is bunk !

that's what i was thinking too.... kinda leary, and i only have 20 plates. Don't wanna go usin em all up... till i get some more on hand.

but if the vendor syringe is bunk, they'll be damn sure to be getting an e-mail from me.
