sterilization i have read a few articles on using pressure cookers to sterilize equipment, and the time it should take to kill all bacteria and what not. Ive also read the cautions about over cooking the medium. all the pressure cookers mention using 10 to 12 psi for a certain amount of time. i happen to have an autoclave at my disposal that hits 20 to 30 psi, and gets up to 250 to 270 degrees. on a warmed up autoclave how long should i sterilize my substrate?

correction 15 psi+ is for all mycological uses. times should be relativley the same. the endospores and what not have to be exposed to the heat for at least 1 hour. at 15psi i still sterilize for 1 and a half hours everytime.

so are you saying not to use the autoclave because to pressure gets too high?

well if your container is strong enough to handle it, it will be fine but some glass jars might not be able to handle the pressure and break inside the cooker.

Incorrect. The holes in your jars will allow pressure to equalize. The pressure will be the same inside your jar as outside. Higher pressure is great. Use it if you got it. Leave the vent or regulator on your autoclave/pc open for a few minutes to exhaust the air in the chamber. Close the vent and allow pressure to build. Give an hour at 20 to 30 psi. I also use an hour and a half at 15psi. This will not overcook your grains. RR
