I'm now on my 4th successful go on this thanks to everyones help here.
The last 2 times I've noticed I may of over pressure cooked my jars and colonization seem to take WAY longer my first go which was not over pressure cooked.
Now instead of 45mins-60mins of pressure cooking I did the instructed 30mins. The jars definitely seem moister than the ones that took a long time to colonization
So.. Now that I followed the directions and my jars seem to be at the correct moisture level will colonization be any faster than if they were dry?

yes jars with the proper moisture level WILL colonise faster then those without adiquate moisture. however jars with to much moistre are prone to infection, rot, and staling so more water isnt allways better but you must have sufficiant water for opimal growth. does all that make sence?

it is all about proper moisture.
if its right, its right if not, not.
concerning the sterilisation times, as for quart jars of grains 60 minutes @ 15psi are what you want, not 30 or anything lower, maybe 2 hours but thats what there is about it.
i autoclaved spawnbags for 3 hours at 20 psi and they went just fine, it sounds as if your problem was the adequate moisture and just that.

grain jars need atleast 60 minutes @ 15psi - in ONE go.

30 mins for Brown Rice flour at 15 PSI should be fine. .. Grain is about double that time
