Hi ppl, yesterday I was talking with some guy at the university and they suggest using jelly without taste and color has solid cultivation medium... what do u think about it? has anyone have experience with this??
also, I'm thinking to had some glucose to the mix, so it has more sugar...
btw, how much glucose do you use in the liquid cultivating medium??
thx for ur opinions ;D

Sounds like a good way to have contaminations in it? Agar only works when it's completely contaminated free, it would probably be easier to go with that.

hum.... "Sounds like a good way to have contaminations in it?"
why u say that??
maybe u didn't understand... see, the jelly is a powder i use 8g of jelly and had 5g of Glucose to 250ml of hot distilled water. then u mix it well, and put it in a pressure cocker for 30 min... after that, en a clear glove box, u set the solution in petri dishes and wait until its get like gel...
I'm just asking if the nutritional part its ok...
the Nutritional Info of jelly (for 100g of powder): Energy(kcal): 372 Proteins: 93g Total fat: 0 Available carbohydrates: 0 Sweeten total: 0

sounds good to me. the stuff is used in some labs for certain purposes as an easy substitute for agar. there's different types though - gelatin made from animal products, and pectin which is made from veggie products (should work very similar to agar) so i'd lean towards pectin if possible.
only concerns would be that gelatin if boiled for too long or too hot (in the case of a PC) damages the protiens and you might lose its ability to gel up after cooling. there's also a concern with incubating temps, it might "melt" back into liquid if you incubate too high, not sure how it will stand up to the mid 80F's that we use. i think these are the reasons agar is preferred as it seems to be pretty "sturdy"
its commonly accepted that you can sterilize plain water with a simple boil at 15 minutes. if it fails in the PC you might want to try just a boil for somewhere between 15-30 minutes.
its worth a shot, might take a few tries to get it right, but you can certainly experiment with "blank" plates to see how it stands up to those conditions before innoculating with anything. somebody asked the same question the other day, so post your results if you find a recipe/method that works out, i'm sure it would be appreciated!

ok, my product is gelatin (just but it at the supermarket), i said jelly because i didn't know the right translation =P
i don't think gelatin melts down, i haven't try it... but, it seams to be a chemical reaction, like with cement...
anyway, maybe you can let it transform in gel, and after that set it in the PC... i will try...
bad enough, i don't have so much money to buy glass petri dishes T_T this research will go sloooooooowwww =P
any other information or advice will be appreciated...

it will melt back down... ever leave a bowl of jell-o in the sun? incubating temps probably aren't going to be high enough to cause that. putting it back into the PC after it gels up almost certainly will destroy the protiens and it won't gel again.
here's an article about how gelatin works might give some insight. those "polypeptide chains" it speaks of will be damaged by excessive heat, making the stuff unable to gel up again.

jajajaja... yep, u hare right... if u set gelatin, after it gels, in the PC, it becomes liquid for ever =P

finally, some test tubes did had jelly in the bottom... that meas that its still possible to get gel
