Cake Questions
    #5533055 - 04/19/06 10:19 AM (17 years, 9 months ago)

Anybody know which method of inoculation is preferrable? I've read through a couple different one's; some say to hold the needle above the cake, and squirt it on the side. I've also heard some people put the needle directly into the cake to inoculate it. Which method promotes faster colonization/lower risk of contamination? Also, should I put a layer of dry vermiculite over top of my mixed cake, and if so, should I separate the two layers with tinfoil?

my suggestion is that you go back and read the pf-tek man. You dont seem to know exactly what your doing and that spells failure with mycology. In your jars you are gonna want your brf and verm mixture with about another layer of dry verm right on top of it. When innoculating you are gonna want to stick the needle down into the jars and just angle it a bit towards the wall of the jar and squirt a bit on each side.

Thanks for clarifying that up carl. Should I seperate the dry verm from the BR mixtures with a layer of tinfoil?? Also, since the dry vermiculite is on top, my jars holes don't need to be taped/tinfoiled or tyvek'd correct? I can just leave the holes open? Is it safer(lower risk of contams) to use tyvek or a coffee filter over the top anyways?

no tinfoil tinfoil does not breath. this would defeat having a dry verm layer. i have yet to see a pf tek mention any tinfoil layer.

you can leave the jar holes open however if you already have tyvek kicking around why not lower the contam risk more i say. personally i use microspore tape but then again i rarely ever do cakes now.

coffee filter is not a substitute for tyvek or microspore tape. its just a dust cover. its not nearly fine enough to trap spores.

when doing cakes you want to always use a top vermiculite layer. this layer helps in keeping contaminates out and retains moisture for later use. don't seperate the vermiculite and the cake, just place the vermiculite on top of the BRF mixture.

for innoculations make sure the needle is all the way into the BRF mixture, past the vermiculite top layer. when you see the needle on the side of the glass reach the BRF mixture, then inect a quarter of a MilLiliter of spore suspention fluid or enough so that you can see a bead of water drip down to the bottom of the jar. this way you know your spores have reached the substrate and are going to germinate. don't inject too much or you could throw off the water content of the jar.

the vermiculite top layer helps keep contaminates out because it containes no nutrients and only water. with no nutrients nothing will germinate on it but the mycelium will grow through it.

hope this helped. ~thenewguy

I use tin foil, but i keep it loose enough to have gas exchange.
Also when inoculating I add another hole in the middle it helps the cake finish colonizing faster.
BTW i don't use jars, i use ordinary glass cups covered with tow layers of foil.
so...just try and see for yourself, after a few times you'll have your own method of doing it...

corrction top layer works beucase it contains no nutes AND no water. a wet top verm layer wouldn't work as well.
