Lids on right-side-up for Rye, bad?

Hello. I pressure cooked some 1 quart ryes with the lid on normal rubber side down (band untightended a little)with tape over the holes... Could this cause a longer-term problem that leads to non-colonization? Thanks!

Rubber side down is good, but no gas exchange isn't going to help anything. Look into drilling one 1/4" hole in the top, stuffed with polyfill, and you can inoculate directly through that--no taping necessary. Then, cover with a coffee filter.
Just my .02, I get quart jars to colonize in 5 days with this setup (using liquid culture).

And i pressure cooked with foil + tape over holes... took out... took off foil + tape and innocutated, then put 2 coffee filters w/ a rubberband over the top (should have optimal air flow). At least now i know it shouldnt make a problem during hte PC'ing. Thanks!
Btw: 5 days, eh? Its been about 3 days with 0 signs of growth... using "from syrninge to rye" he had mad growth after 3 1/2 days... Is that uncommon? How long should one wait before giving up on a quart of inoculated but uncolonized rye? I used 2.5cc per jar of syrninge from Lil Shop of Spores (the syringes looked a little shady compared to the darker ones from Spore Works, so i donno, worked well on some 1/2 pints with other strains from them).

I hope your using Micropore over those holes as that will work perfect for gas exchange. If you covered your basics such as shaking the syringe prior to inoculating and incubation temps around 84-86F. to speed up the process you'll be just fine but you'll need to pick up some patience if you get a chance. Spores can take their own sweet time sometimes to germinate I've seen up to 3 weeks even personally. GL

Sounds good... yea... been anxious trying differeent methods. hehehe. I guess i will make more jars w/ diff substrate as if these are lost... but still see how the end up... worse comes to worse, i have a lotta coolonized substrate that will come to good use Patience... definately need to pick some of that up.
