Those puertos-- birdseed and partial sterilization

Came home to a happy sight today.

I made up some jars with Ryche's birdseed recipe last week, only to discover that my roommate had completely fucked my pressure cooker (tried to cooked some beans with it and let the water boil away, thereby riasing the temperature too high which then melted the alloy pellet in the safety valve...). I had read in one of Anno's posts that it is possible to sterilize grain via multiple boilings... so I figured what the hell. While I was waiting for my new pressure cooker to be delivered... I decided to try out the partial sterilization method.

I boiled the jars for one hour the first day, waited 24 hours, boiled again, waited 48 hours and put them in the oven at 250 degrees, and then to be safe, boiled them again a day later. I opened one jar to see if the substrate was still moist enough.. and it was. I tossed its contents and got to work on the other jars.

So, I went ahead made up 7 syringes with my puerto print and used two of them to innoculate the jars and placed them in my nifty incubation chamber on Saturday night. I saw nothing at all except the tiniest pin-sized signs of growth last night. I came home today, and the jars went buck wild... one of them is 1/3 covered with mycellium.

These babies grow fast... also, no signs of molds or greasy bacteria contams yet, and I smell nothing through the filter disks that indicates anything undesirable cohabitants in the jars.

I will post further details as they develop. If these jars do not fall victim to contams, it would be good evidence that partial sterilization can work with birdseed subtrates, which might bode well for those who cannot afford a PC but want to try non BRF subtrates.

Good to hear of your apparent success.

Fractional sterilisation works, I tried it myself not so long ago.

I followed this standard procedure, except that I boiled the jars for 45 minutes at the lat boil:

Tyndallization (fractional sterilisation) process

1) Steam heating to 90-100 ?C for 30 min
Vegetative cells are destroyed but endospores survive

2) Incubate at 37 ?C overnight
Most spores germinate

3) Second heat treatment 90-100 ?C, 30 min
Germinated spores are killed

4) Second incubation at 37 ?C overnight
Remaining spores germinate

5) Third heat treatment 90-100 ?C, 30 min
Germinated spores are killed
