Why no mycelium growth?
    #5457628 - 03/29/06 11:03 PM (17 years, 9 months ago)

Well this is my first time using wild bird seed. Did PF cakes before this in 1 pint jars. I bought bigger quart jars for the WBS. Here's what I did.
Went to PetsMart and bought a 40lb. bag of basics wild bird seed. ($8)
Soaked half of it overnight in a 5 gallon bucket. (10 hours)
Before I strained the WBS after the soak, I mixed it up so most of the sun flowers would float on top, and took out 90% of them. Then strained for 30+ minutes and loaded into 1 quart jars 2/3 full.
Drilled a hole in the lids for the pollyfil and put 2 coffee filters on each jar for an extra filter.
Then PCed at 15psi for 90 minutes.
After it cooled down overnight, I used the oven tek to inoculate. Some people say its bad, but last time I used it to inoculate my PF cakes, out of 24 I had 1 contaminated.
Took off the coffee filters and stuck the needle through the pollyfil so I wouldn't even have to expose it to the contaminated air, wiped the lids wit alcohol before replacing the coffee filters. Between each jar I flamed the needle and kept it in an alcohol soaked cotton ball. I have 2 jars each of :
P.E.S Hawaiian
Blue Meanie
Golden Teacher
Treasure Coast
Texas
Pink Buffalo
Each jar got shooken up after inoculation and placed in an incubator.

Now its been 6 days and there is no sign of mycelium growth. One of the jars has 3 tiny spots, but other then that can't see anything.
Did I do something wrong? Was I not supposed to shake it after inoculation?? Or did I miss something when preparing the WBS?

EDIT: I just inspected the jars, and all but 1 have small spots with mycelium growth. Maybe they're just off to a slow start.

A 10 hour "cold" soak isn't long enough for them to hydrate properly. I soak for 24 hours and then simmer them for a bit. My driest jars were a 24 hour soak followed by a 1 hour soak in hot water. Some do just a 24 hour cold soak, I wouldn't recommend less.

You might consider injecting some sterile water, but I don't have the experience to tell you how much more water might be good for that level of hydration.

Then again, if you're seeing growth, all may not be lost (though 6 days is way to long to for just specks in a single jar). Compare some dry seeds to the ones in the jar. Do the seeds in the jar looked puffed up?

Incubating at 80 degrees. From what I read here it said to soak for 8-10 hours, and if you let em soak for a whole day, they may not be useable anymore.
But next time I will do a soak for 24 hours and see what the results are. And is LC "hard"? I haven't read much but it does look a bit complicated.

PS: Just picked some Equadors from my PF cake casings, yummy.

I think shaking your jars right after inoculation is your "problem" - but I use the term loosely.  The fact that you see some small spots means you're probably going to be ok.  Because you shook it up your spores got distributed throughout, and you very well might have growth going on on the insides where you can't see.  Not a bad thing IMO from a colonization standpoint, there's probably lots of little spots taking off inside... from an impatient-and-wanting-to-see-something standpoint its a "problem."  Innoculating against the glass wall on the outer edges and not shaking right away only really serves the purpose of giving you that quick indication at the edge of the jar that something is happening.  Give it a few more days and I have a feeling you'll be pleasantly surprised

Oh and P.S. - LC is actually pretty easy.  I reccomend the honeywater tek... don't even need to PC, have gotten perfect results just steaming in a pot for 45 minutes.  I've seen white fluffy growth in the liquid in just over 48h!  Not only will a LC "multiply" your inoculation liquid, but you effectively shave 4-5 days off colonization times when you innoc with myc water vs. straight spores.  Nice to keep a few jars on hand in the fridge as they keep for some time!
