Homemade LC Jars
    #5451907 - 03/28/06 04:25 PM (17 years, 10 months ago)

I've read that you can make your own and i've done it before but I had a Q :P

I put two holes...one for gas exchange (polyfil in that) and another smaller one for injection (siliconed it)

Now should I put a layer of tyvek under the lid? I figure that when you shake it, the polyfil will get wet, which is BAD...lol..So I figured some tyvek would help protect the polyfil from getting wet..

Also...after I stick the needle in, it'll pierce the tyvek and make a hole which will leak into the part where the polyfil is, which is bad as well..so...what Should I do about this?

EDIT ----

Maybe I could put some silicone between the lid and the tyvek (where the needle would pierce), perhaps that would help...Hmmmmm

Anyone use a different method? If not, i'll just go with this.

TIA

I put some broken glass in the jar, this way you can just swirl it and it wont get teh polyfill wet.

I suppose that would work, but I'm new, so my experience is pretty nil. I made my own LC's using agars LC tek, with tubing type gas exchange filters.

It was super simple and quick, and is working great so far. I found the materials with little effort, and it was ready in no time at all.

Search the Karo TECH, afoaf followed the instructions and things went as planned.

I use plastic quart jar lids with poly-fill in one hole and a 1/2" hole drilled with a vacutainer injection port. The 1/2" is a perfect fit for the hole and requires no sealing. I personally use magnetic stir bars with a stir plate for agitation, so I can't be much help on your application.

The issue I see with shaking an LC and getting your Tyvek wet is that you may ruin their porous properties. If the LC gets it wet and drys on it, I don't see how it can be porous again.

Here is a pic of my lid:

I know what tek you're talking about and i'm pretty sure it was the one that is simplified(sp?)...As in it doesn't go through steps are detailed as what i'm looking for.

I used the search function and as always, it drove me insane and brought up Crap

But I think i've devised a way of using silicone that will solve my problem.

Doesn't sound like to bad of an idea. However i'd have to agiate the solution enough to get the glass moving and to do that i'd have to shae it quite a bit, i.e, i'd get the polyfil wet after I stick the needle in...but thanks tho

Exactly...Damnit...Wish I had a stir plate, hahah. Oh well.

I'll do what I can. Nice lid btw, reminds me of Agar's kind of.

I decided to go ahead and just put on Tyvek, metal lid...two holes, one with poly fil and one with silicone...I made it to where the silicone went all the way through the lid and into the tyvek part...

This way the hole in the tyvek will close up

So all should go well.

Thanks everyone for the help tho!

I don't even bother agitating my LCs. A slight swirl maybe to get things moving, but it's nothing major. Those are all great ideas if you care to agitate them tho. I just let them sit for a little longer and they get done. You should try just letting them be and see how that goes for you. GL.

I'd avoid glass chunks and would go with a stubby 1/2" stainless bolt instead. This way you can gently swirl the liquid medium and the bolt somewhat stays put. This breaks up the mycelium very well. Much better than using shards of glass that swirl along with the medium, defeating the purpose.

Indeed, but I don't want it to clump, when it clumps it becomes a mass and really thick, and can't be sucked up into syringes. So I'd like to swirl it every now and then...And I'd like to really break it up towards whenever syringes are to be made.

Sounds good. The threads would catch the mycelium well w/o even having to swirl it to hard. damn , why didn't I think of that, hahah.

Thanks man.

And so ends this thread :P

Thanks for the help everyone, tc
