help diagnose problem

i prepared the original pf cake recepie as sterile and best as i could and then i boiled the finished sealed jars some number of hours and let cool over night. i inoculated the next day and that was 2/9, it is now like 3/22 or whatever and i checked the cakes and only one grew micelium and only over about 33 precent of the jar

what gives? i think i may have killed the fungus cause i wanted to speed colonization so i put a heating pad under the jars and covered the whole thing with a cloth, so temps may have gotten in the 90-100 degree range. only other possibility is too little mosture, can someone explain what a cake should feel like to the touch just after inoculation? i think i may open one and try to see how much moisture is imbedded in the cake. otherwise i can't figure out why most didn't colonize?

also i bought some grain jars from an online shop and inoculated them liberally and put them under the same heating pad and these also did not colonize at all, i shook them 1 week after inoculation bust since have not looked at them until now. I bought the b+ strain from lil shop of spores, i'd suspect the syringe wasn't legit but i do indeed see mycelium on 33% of 1 of about 15 cake jars

Stalled growth like that might have something to do with them not having enough oxygen. It is hard to believe that they would not even germinate though without it... Incubation really helps speed up the growth, but it isn't necessary and if you can't build a proper incubator, you should just let them colonize at room temperature.

If you open one just make sure it is moist. If it is bone dry it is too dry, but even if it is a little wet it probably isnt the reason none of them grew.

i'd bet you fried them with your heating pad. I agree with porkSoda. If you can't build a proper incubator then don't build one at all. You'll probably burn your house down otherwise.

And if the moisture was a problem, then you may find that the center of the cake is colonized when you open it up, meaning that they germinated, but didn't have enough water to go on...

Did you shake your syringe like a madman? Also heating pads get much higher than 90-100 degrees remember your body temp is 98.6 bro! You need to follow a tek to the "T" if you want to succeed. GL

thanks to you all for the responses. for the record, anyone know what heat range will cause death? i was under the impression it was around 105.

I have pretty much 15 cakes looking as though they were just sterilized, think i should go ahead and re-inoculate and let them sit at room temp and wait another month?

i'll be sure to open one cake and check the moisture level and i'll also shake the syrince to disperse the remaining

I don't know the exact figure, but anything over about 85 can't help the mycelium at all and may invite contams to join the party. If I were you, I'd boil them again. Can't hurt, right? :smile:
